摘要
微塑料能在海洋生物体内富集,对海洋生物和环境的污染日益严重。为检测南海乌贼体内微塑料富集情况,以虎斑乌贼为对象,分析其组织中微塑料的富集情况。采用HNO3/H2O2组合液、75℃磁力搅拌消解乌贼鳃、胃及其内容物、肌肉组织,消解液进行氯化钠密度分离,上清经玻璃纤维滤膜抽滤,烘干滤膜,体式显微镜检测。结果表明:HNO3/H2O2消解液、75℃消解条件,使微塑料回收率高于96.29%,对塑料腐蚀性小,为有效消解方法,采用该种方法,在乌贼鳃、胃及其内容物、肌肉组织中检测到红色、蓝色、黑色纤维状微塑料,长条形黄色片状微塑料,块状微塑料。乌贼组织内检测到微塑料,水体中微塑料可能富集在生物体内,通过食物链传递,对其它营养级生物健康存在未知风险。
Microplastics can be enriched in marine organisms.The pollution of microplastics to marine environment and biology is increasingly serious.In order to detect the accumulation of microplastics in the cuttlefish in the South China Sea,the Sepia pharaonis was selected as the research object,and the enrichments of microplastics in the tissues of Sepia pharaonis were analyzed.The gill,stomach and its contents,and muscle tissue of cuttlefish were digested by HNO3/H2O2 combined solution and 75℃magnetic stirring.The digested solution was separated by sodium chloride density.The supernatant was filtered by glass fiber filter membrane,the filter membrane was dried and detected by body microscope.The results showed that HNO3/H2O2 digestion solution and 75℃digestion condition is an effective digestion method,which make the recovery rate of plastics higher than 96.29%and make the plastics less corrosive.Using this method,red,blue and black fibrous microplastics,long yellow sheet microplastics and block microplastics were detected in the gills,stomach and contents,and muscle tissues of cuttlefish.Microplastics are detected in squid tissues.Microplastics in water may accumulate in organisms and pass through the food chain,which may pose unknown risks to health of other nutritive organisms.
