基于GTEx数据库对COVID-19结合基因ACE2的分析 被引量:2

摘要 目的探讨血管紧张素转化酶2受体基因(ACE2)在正常人组织器官中的表达,结合新型冠状病毒肺炎(COVID-19)患者临床症状分析两者间的关系,利用生物信息学方法揭示ACE2可能的效应通路。方法从基因型-组织表达(GTEx)数据库下载ACE2在人体各组织的表达数据,并用R软件对数据进行处理。通过GO富集分析其相关机制,并探讨长链非编码RNA和ACE2的相关性。结果ACE2在正常人体不同器官均有一定程度的表达,表达量从高到低依次为:心、肾、肺、肝、胃、脑、脾,差异有统计学意义(F=820.03,P<0.001)。ACE2在不同组织中也有一定程度的表达,表达量从高到低依次为:睾丸、脂肪、神经、肌肉、血,差异有统计学意义(F=3592.75,P<0.001)。GO富集结果提示,基因ACE2主要参与调节肽和激素的分泌、调节蛋白质异二聚活性;同时长链非编码LINCO2082与ACE2成显著正相关性。结论ACE2在人体各组织器官均有一定程度的表达,可能与COVID-19患者器官损害存在一定的相关性,临床应密切观察。 Objective To determine the expression of angiotensin converting enzyme 2 receptor gene(ACE2)in human normal tissues and organs and analyze the correlation between ACE2 expression and clinical symptoms of COVID-19 patients.The potential effect pathways of ACE2 were disclosed with bioinformatics analysis.Methods The data of ACE2 in human tissues were collected from the database of Genotype-Tissue Expression(GTEx)and R software was used to interpret these data.The mechanism of ACE2 was analyzed by GO enrichment and its correlation with long non-coding RNA was also studied.Results ACE2 was expressed in different human organs.The expression levels from high to low were:the heart,the kidney,the lung,the liver,the stomach,the brain,and the spleen,with significant difference(F=820.03,P<0.001).ACE2 was also expressed in different tissues.The expression levels from high to low were:the testis,fat,nerve,muscle,the blood,and the difference was statistically significant(F=3592.75,P<0.001).GO enrichment showed that ACE2 gene was mainly involved in the regulation of peptide secretion,and protein heterodimerization activity.Meanwhile the long non-coding RNA 2082(LINCO2082)had a significant positive correlation with ACE2.Conclusion ACE2 is expressed in various human stissues and organs,which may be associated with the organ impairment in COVID-19 patients.More attention should be paid to these organs in clinical care.
