维生素B2修饰的纳米酶对紫外线诱导HaCaT细胞氧化应激损伤的作用及机制

摘要 目的探讨维生素B2修饰的纳米酶(VB2-IONzymes)对中波紫外线(UVB)照射后HaCaT细胞氧化应激损伤的作用及可能的机制。方法UVB照射诱导HaCaT细胞光损伤模型,将培养的HaCaT细胞分为空白组、UVB照射组、VB2-IONzymes组、UVB+VB2-IONzymes组。CCK-8检测HaCaT细胞增殖活性,酶生化法检测细胞醛缩酶(MDA)及超氧化物歧化酶(superoxide dismutase,SOD)水平,ELISA法检测细胞中Nrf2及HO-1蛋白水平。结果UVB照射组HaCaT细胞增殖活性显著降低,细胞MDA水平升高,SOD水平降低,Nrf2及HO-1蛋白表达下降,与空白组相比,差异均有统计学意义(P<0.01);加入VB2-IONzymes后,与UVB照射组相比,细胞活力明显提高(P<0.05),细胞MDA水平显著降低,细胞内SOD水平、Nrf2及HO-1蛋白表达均显著上升(P均<0.01)。结论VB2-IONzymes有抗氧化应激作用,其机制可能与促进Nrf2和HO-1表达相关。 Objective To explore the effect and mechanism of vitamin B2 modified IONzymes(VB2-IONzymes)against oxidative stress injury in HaCaT cells after ultraviolet irradiation.Methods The light injury model of HaCaT cells was induced by UVB irradiation.The HaCaT cells were divided into untreated control group,UVB irradiation group,VB2-IONzymes group and UVB+VB2-IONzymes group.The proliferation of HaCaT cells was detected by CCK-8,MDA and SOD were measured by enzyme biochemical method,the expression of Nrf2 protein and HO-1 protein was determined by ELISA assay.Results The viability of HaCaT cells was decreased after UVB irradiation,the MDA level was decreased,the SOD level,the expression of Nrf2 protein and HO-1 were up-regulated when compared with the blank group,and the differences were significant(all P<0.01).After treated with VB2-IONzymes,the viability of cells in UVB+VB2-IONzymes group was increased significantly(P<0.05).MDA level was decreased,SOD,Nrf2 and HO-1 protein expression were increased compared with UVB irradiation group(all P<0.01).Conclusion VB2-IONzymes has the effect of antioxidant stress,which may through increasing the expression of Nrf2 and HO-1.
