摘要 目的:探讨样本采集、运输和储存过程,对HIV病毒核酸检测结果的影响。方法:选取2019年4-8月检验科接收HIV阳性样本5份,使用阴性浆对标准本进行梯度稀释,使用EASYQ 2.0 HIV-1核酸检测试剂检测,模拟不同采集操作、不同储存时间和不同溶血程度,分析试剂检测灵敏度。结果:EASYQ 2.0 HIV-1核酸检测试剂盒最高灵敏度可达10 Cps/mL,4℃下保存与室温保存可能对样本检测结果产生影响,溶血是影响HIV-RNA检测的最大风险因素。结论:样本采集、运输和储存过程都可能影响HIV病毒核酸检测结果,严格按照标准流程操作,能减少样本报废率,提高检测结果准确性。 Objective:To investigate the impact of sample collection,transportation and storage on the results of HIV nucleic acid testing.Methods:5 HIV positive samples received by the laboratory from April 2019 to August 2019 were selected.The standard was diluted with negative plasma in gradient.EASYQ 2.0 HIV-1 nucleic acid detection reagent was used to simulate different collection operations,different storage time and different degree of hemolysis,and the sensitivity of the reagent was analyzed.Results:The highest sensitivity of the EASYQ 2.0 HIV-1 nucleic acid detection kit was 10 Cps/mL.Storage at 4℃and room temperature may affect the test results.Hemolysis is the biggest risk factor affecting HIV-RNA detection.Conclusion:The process of sample collection,transportation and storage may affect the results of HIV nucleic acid testing.Operating strictly in accordance with standard procedures can reduce the rate of sample rejection and improve the accuracy of test results.
