三相萃取法分离纯化竹黄多糖工艺优化 被引量:2

摘要 采用三相萃取法纯化竹黄菌(Shiraia bambusicola P.Henn.)多糖,考察叔丁醇-(NH4)2SO4-水提液体系的三相萃取性质,研究提取液中(NH4)2SO4的质量分数、叔丁醇添加量、萃取温度对竹黄提取液多糖萃取率和蛋白质去除率的影响。在单因素试验的基础上,通过Box-Benhnken组合设计和响应面分析法优化三相萃取的最优工艺。响应面分析结果表明,竹黄子实体中多糖的最佳提取条件为:萃取温度35.16℃,叔丁醇∶样液体积比0.96∶1,硫酸铵质量分数26.10%,此条件下的多糖萃取率为90.43%,蛋白质去除率可达88.45%,该体系是一种可行的高效分离纯化竹黄多糖的绿色方法。 Shiraia bambusicola polysaccharides were purified by the three phase extraction method.and the three phase extraction properties of tertiary butanol-(NH4)2 SO4-aqueous extract system were investigated.The effects of three factors,including the amount of(NH4)2 SO4 in the extract,the amount of tertiary butanol and temperature of the extract on the extraction rate of polysaccharides and protein from Shiraia bambusicola extract were studied,and the extraction conditions of polysaccharides were optimized by Box-Behnken experiment design and response surface analysis.The optimum extraction conditions were as follows:extraction temperature 35.16℃,ratio of t-butyl alcohol and crude extract 0.96∶1(V·V-1)and ammonium sulfate concentration 26.10%.Under this condition,the polysaccharide extraction rate was 90.43%and the protein removal rate was88.45%.Therefore,the system would be a feasible and efficient method for separating and purifying Shiraia bambusicola polysaccharides.
