昼夜节律对动脉粥样硬化凝血系统的影响 被引量:1

Effect of Circadian Rhythm on Atherosclerosis Hemostasis System

摘要 目的研究凝血系统中具有昼夜节律性的基因,分析其在冠状动脉粥样硬化中的调控作用。方法通过选取人循环血液表达谱的相关数据,利用LSPR分析其中基因表达的昼夜节律性,利用Reactome数据库凝血通路及CirGRDB找出具有昼夜节律性的凝血基因,分析动脉粥样硬化相关基因,研究昼夜节律性凝血因子具体凝血功能及在动脉粥样硬化病变中作用。结果具有昼夜节律性的相关凝血基因功能为血小板止血,血小板激活和聚集,纤维蛋白凝块级联反应形成,纤维蛋白凝块溶解,细胞表面与血管壁的相互作用,巨核细胞发育与血小板生成的相关因子,部分凝血因子与不稳定冠状动脉粥样硬化斑块相关,昼夜节律干扰后相关节律性减弱或消失。结论部分凝血因子具有昼夜节律性,动脉粥样硬化斑块中也高表达,节律紊乱干扰相关凝血因子节律性。 Objective To study genes in the hemostasis system that have circadian rhythms,and to analyze its regulatory effect in coronary atherosclerosis.Methods Human circulating blood expression spectrum was collected by LSPR to analyze the circadian rhythm of gene expression.Reactome database hemostasis pathway and CirGRDB were used to find the hemostasis genes with circadian rhythm and analyze the genes associated with atherosclerosis.The specific hemostasis function of circadian hemostasis factor and its role in atherosclerosis lesions were investigated.Results Circadian rhythmatic hemostasis genes functions in platelet hemostasis,platelet activation signaling and aggregation,formation of fibrin clot cascade,dissolution of fibrin clot,cell surface interactions at the vascular wall,factors involved in megakaryocyte development and platelet production,part of the hemostasis factors associated with unstable coronary atherosclerotic plaques,circadian rhythm were impair or disappear after the rhythm disturbance.Conclusion Part of the hemostasis factors appear circadian rhythm,also high expressed in atherosclerosis plaque,rhythm disturbance interferes with the rhythm of coagulation factors.
