水飞蓟素调控信号传导及转录活化子3逆转乳腺癌耐药性的研究 被引量:6

摘要 目的探讨水飞蓟素是否可通过调控STAT3的表达逆转乳腺癌的耐药性。方法通过Western Blotting和QPCR实验检测乳腺癌亲本细胞株MCF⁃7和耐药株MCF⁃7/MDR中STAT3的表达;通过转染STAT3小干扰RNA(si⁃STAT3)和STAT3质粒(STAT3⁃OE)分别下调和上调耐药株中STAT3的表达;CCK⁃8和流式细胞术检测水飞蓟素和STAT3对MCF⁃7/MDR细胞中阿霉素毒性和细胞凋亡能力的影响。结果MCF⁃7/MDR中STAT3的表达水平较亲本细胞株MCF⁃7提高;水飞蓟素处理可降低MCF⁃7/MDR细胞的IC50值,促进细胞凋亡;STAT3可增加IC50值,抑制细胞凋亡;同水飞蓟素组相比,水飞蓟素+STAT3⁃OE组IC50值提高,凋亡水平降低;同si⁃STAT3组相比,水飞蓟素+si⁃STAT3组细胞的IC50值无明显变化。结论水飞蓟素可通过降低STAT3的表达逆转乳腺癌耐药性,可为临床上乳腺癌的治疗提供新的思路。
