减数分裂恢复前期小鼠卵巢组织中M-CSF和NPPC的表达曲线

摘要 目的通过建立促排卵小鼠模型,研究减数分裂恢复前期小鼠卵巢组织中巨噬细胞集落刺激因子(M-CSF)和其受体(M-CSFR)、C型尿钠肽前体(NPPC)及其编码的C型尿钠肽(CNP)表达曲线及其相互关系。方法建立促排卵小鼠模型,在人绒毛膜促性腺激素(hCG)促排卵后0、0.5、1、2、4 h分别以免疫组化和RT-PCR方法检测卵巢组织中M-CSF、M-CSFR、NPPC蛋白及mRNA的表达水平。结果促排卵后,M-CSF、M-CSFR蛋白及mRNA表达水平均呈下降趋势,而NPPC mRNA表达水平呈上升趋势,并在1 h达峰值后显著下降,CNP表达水平在2 h上升至峰值后,随即快速下降至较低水平。结论排卵前促黄体生成素峰后,卵巢组织中M-CSF及其受体表达显著下降,引起一系列信号传导,导致CNP表达水平在2 h达到峰值,随即快速下降至较低水平,最终卵母细胞减数分裂恢复及排卵。 Objective To generate the expression curves of macrophage colony-stimulating factor(M-CSF)and its receptor(M-CSFR),natriuretic peptide precursor type C(NPPC)and C-type natriuretic peptide(CNP)in mouse ovaries during meiotic resumption.Methods Ovaries obtained from the ovulation-induced mice at 0 h,0.5 h,1 h,2 h and 4 h after the hCG injection were used to analyze the expression of M-CSF,M-CSFR and NPPC by both immunohistochemistry and quantitative fluorescence PCR.Results After human chorionic gonadotophin(hCG)injection,the protein and mRNA expression of M-CSF and M-CSFR decreased.The expression levels of NPPC mRNA increased after hCG injection,reached the peak at 1h and then decreased significantly.The expression of NPPC-encoded protein CNP reached the peak level at 2 h after hCG injection and then decreased rapidly to a lower level.Conclusion The pre-ovulatory luteinizing hormone(LH)/hCG surge decreases the expression of M-CSF and M-CSFR in the ovaries,which caused a series of signal transduction,leading to the peak of the CNP level at 2 h,then decreased rapidly to a lower level,and finally causing the meiosis resumption and ovulation.
