消肿散对大鼠急性软组织损伤PGE2、IL-6及IL-1β影响的实验研究 被引量:4

Experimental Study on Effect of Xiaozhong Powder on PGE2, IL-6 and IL-1β in Rats with Acute Soft Tissue Injury

摘要 目的本研究通过动态观察消肿散对大鼠血清中炎症因子PGE2、IL-6和IL-1β的表达量的影响,探讨该方治疗本病的作用机制。方法首先从50只大鼠中随机选出5只作为空白组,其余大鼠造模,造模后从中选出5只作为模型组进行模型鉴定。鉴定完成后将剩余的40只大鼠随机分为消肿散组和模型组,然后将这2组再随机分为4个小组:1 d组、3 d组、6 d组和10 d组,每个小组5只。给予消肿散组大鼠消肿散外敷,模型组生理盐水外敷,将2组大鼠于以上时相点分批处死并制作标本。用ELISA法检测血清中PGE2、IL-6和IL-1β的含量,最后统计分析数据。结果消肿散组与模型组大鼠血清中PGE2、IL-6和IL-1β的表达量均先上升,在第3天达到峰值,后下降直至第10天。干预1 d后,2组大鼠血清中以上炎症因子的表达量比较差异有统计学意义(P<0.05或P<0.01);干预3 d后,与1 d组比较,2组大鼠血清中以上炎症因子的表达量均上升,且消肿散组低于模型组,两组比较差异有统计学意义(P<0.01);干预7、10 d后,2组均有明显降低,且消肿散组低于模型组,2组比较差异有统计学意义(P<0.01)。结论消肿散所具有的消肿、镇痛等治疗作用,与其能降低血清中PGE2、IL-6和IL-1β等炎性因子的表达量有关,本课题为消肿散在临床推广运用提供了一定的科学依据。 Objective In this study,the effect of Xiaozhong San on the expression of inflammatory factors PGE2,IL-6 and IL-1βin serum was observed by dynamic observation.Methods Firstly,5 out of 50 rats were randomly selected as blank group,and the other rats were modeled.Five models were selected as model groups for model identification.After the completion of the identification,the remaining 40 rats were randomly divided into the ablation group and the model group,and then the two groups were randomly divided into 4 groups:1d group,3d group,6d group and 10d group,each group 5 only.The rats in the Xiaozhong powder group were given external application of Xiaozhong powder.The rats in the model group were treated with normal saline.The rats in the two groups were sacrificed and specimens at the above time points.The levels of PGE2,IL-6 and IL-1βin serum were detected by ELISA,and finally the statistical data were analyzed.Results The expression levels of PGE2,IL-6 and IL-1βin the serum of Xiaozhong San group and model group increased first,reached the peak on the third day,and then decreased to the 10th day.After 1 day of intervention,the expression levels of inflammatory factors in the serum of the two groups were statistically significant(P<0.05 or P<0.01).After 3 days of intervention,compared with the 1d group,the inflammatory factors in the serum of the two groups were compared.The expression levels increased,and the Xiaozhong San group was lower than the model group.The difference between the two groups was statistically significant(P<0.01).After 7 days and 10 days,the two groups were significantly lower,and the Xiaozhong powder group was lower than the group.In the model group,the difference between the two groups was statistically significant(P<0.01).Conclusion The therapeutic effect of Xiaozhong powder on swelling,analgesia,etc.,is related to the reduction of the expression levels of inflammatory factors such as PGE2,IL-6 and IL-1βin serum.This topic provides scientific basis and a certain contribution to the clinical application of Xiaozhong powder.
