摘要
目的对汉坦病毒三种实验室检测行业标准核酸检测方法进行比较和验证。方法应用三种标准方法检测含有体外合成汉坦病毒核酸的模拟鼠肺样本,对特异性、灵敏性、检测下限、重复性等技术参数进行比较,并选择最优方法进行鼠类监测研究。结果三种标准方法均可检出汉坦病毒核酸,但其中第二种标准方法灵敏度、重复性、检测下限等指标均较好,通过该方法,在多个口岸鼠类监测中多次检出汉坦病毒。结论当存在多种检测方法时,实验室应进行验证和比较,筛选出最合适的方法作为实验室检测的首选方法。
Objective To compare and verify three laboratory standard nucleic acid detection methods for Hantavirus.Methods For the three standards methods,a head-to-head comparison on specificity,sensitivity,lower limit of detection,and reproducibility was conducted using Hantavirus spiked mouse lung samples.The best method was selected for the routing rodent surveillance.Results Hantavirus nucleic acid can be detected by all three standard methods,but the sensitivity,reproducibility,and lower limit of detection of the second standard method are better.Using this method,Hantavirus has been repeatedly detected during routing rodent surveillance in Gongbei port.Conclusions Comparison and verification should be conducted to screen out the best method for laboratory testing when multiple methods exist.
