HOTAIR在甲状腺乳头状癌中的表达及对增殖与侵袭的影响

摘要 目的探讨长链非编码RNA(long non-coding RNA,lncRNA)HOTAIR在甲状腺乳头状癌(PTC)中的表达及其对癌细胞增殖与侵袭的影响。方法采用荧光定量PCR技术检测PTC患者肿瘤组织、血浆中HOTAIR的表达水平,分析其与患者一般临床病理参数间的关系,分析HOTAIR的表达对甲状腺乳头状癌的诊断意义。结果lncRNA HOTAIR表达和PTC患者性别、年龄、瘤体直径等均无关(P>0.05),而与淋巴结转移、TNM分期相关(P<0.05);转染36 h、48 h后,Si-HOTAIR细胞增殖活性显著低于Si-NC,转染pcDNA3.1-HOTAIR细胞增殖活性相比pcDNA3.1提升。Si-HOTAIR和Si-NC对比,Matrigel基底膜的细胞数量显著降低,且pcDNA3.1的细胞数量低于pcDNA3.1-HOTAIR。结论长链非编码RNA HOTAI的检测对甲状腺乳头状癌具有良好的预测作用,有望成为PTC早期诊断的分子标志。 Objective To investigate the expression of long non-coding RNA(lncRNA)HOTAIR in thyroid papillary carcinoma(PTC)and its effect on the proliferation and invasion of cancer cells.Methods The expression levels of HOTAIR in tumor tissues and plasma of PTC patients were detected by fluorescence quantitative PCR,and the relationship between HOTAIR and the general clinicopathological parameters of patients was analyzed,so as to analyze the diagnostic significance of HOTAIR expression in thyroid papillary carcinoma.Results The expression of lncRNA HOTAIR was independent of PTC patients′gender,age,and tumor diameter(P>0.05),but correlated with lymph node metastasis and TNM stage(P<0.05).After transfection for 36 h and 48 h,the proliferation activity of si-hotair cells was significantly lower than that of si-nc cells,and the proliferation activity of pcdna3.1-hotair cells was improved compared with that of pcDNA3.1.Si-hotair compared with si-nc,the number of Matrigel basal membrane cells was significantly reduced,and the number of pcDNA3.1 cells was lower than that of pcdna3.1-hotair.Conclusions The detection of long non-coding RNA HOTAI has a good predictive effect on thyroid papillary carcinoma and is expected to be a molecular marker for early diagnosis of PTC.
