摘要
目的:对比研究马钱子-甘草药对配伍前后及其拆分相态指纹图谱。方法:采用Diamonsil C18(2)(250 mm×4.6 mm,5μm),流动相乙腈-0.1%磷酸溶液梯度洗脱,流速1.0 mL/min,柱温25℃,进样量10μL,检测波长254 nm;获得马钱子单煎液、甘草单煎液、二者合煎液及配伍相态的色谱图,建立合煎液的指纹图谱,标定共有峰,对配伍后指纹图谱的共有峰进行归属分析,对比各拆分相态的色谱峰。结果:通过马钱子-甘草药对的指纹图谱,标定了25个共有峰,5个来源于马钱子,20个来源于甘草,指认出9个成分,配伍前后峰的数目未发生变化。结论:所建立的指纹图谱方法准确、可靠,可较全面的反映了马钱子-甘草配伍后指标成分的特征,配伍前后色谱峰无差异,沉积物相态为配伍相态的关键所在,以上结果可为甘草-马钱的配伍应用和减毒研究提供参考。
Objective:To study the fingerprints of Nux vomica-Glycyrrhiza before and after the compatibility and its split phase fingerprint.Methods:Diamonsil C18(2)(250 mm×4.6 mm,5μm)was applied,gradient elution was carried out with mobile phase of acetonitrile-0.1%phosphoric acid solution,the flow rate was 1.0 ml/min,column temperature was 25℃,sample size was 10μL,detection wavelength was 254 nm.Firstly,the chromatograms of Nux vomica decoction,Glycyrrhiza decoction and the mixed decoction were obtained;on which basis,the fingerprints of the mixed decoction were established,and the common peaks were calibrated.The common peaks of fingerprints after the compatibility were assigned.Results:The fingerprints of the pair of Nux vomica-Glycyrrhiza identified 25 common peaks,5 from Nux vomica and 20 from Glycyrrhiza;9 components were identified.There were no changes in the number of peaks before and after compatibility.Conclusion:There was no difference in the chemical composition of the Nux vomica-Glycyrrhiza before and after the compatibility.The established HPLC fingerprint spectrum fully reflects the characteristics of the index components after the combination of Nux vomica-Glycyrrhiza.The analysis method is accurate,reliable and reproducible.
