融合魏斯氏菌(Weissella confusa)XG-3产胞外多糖条件研究 被引量:9

摘要 以分离自香瓜培养物中产胞外多糖(exopolysaccharide,EPS)乳酸菌(lactic acid bacteria,LAB)融合魏斯氏菌(Weissella confusa)XG-3为供试菌,通过扫描电子显微镜(scanning electron microscope,SEM)观察显微形态。以提高EPS产量为目标,采用单因素试验法优化该菌株产EPS发酵条件。结果表明:培养基组分为葡萄糖90 g/L、酵母提取物10 g/L、乙酸钠6 g/L、柠檬酸铵5 g/L、吐温802 mL及初始pH为6.5,培养条件为摇床转速120 rpm、温度30℃时,W.confusa XG-3 EPS产量达到61.03 g/L,较优化前增加了44.95%。 EPS-producing lactic acid bacteria strain,Weissella confusa XG-3 which was isolated from fermentation culture of Cucumis melo L was used as the test bacteria.The micromorphology of W.confusa XG-3 was observed by SEM.The fermentation conditions of XG-3 EPS was optimized by single factor experiment and the yield of EPS was used as an indicator.The results showed that the composed of optimal culture medium was glucose 90 g/L,yeast extract 10 g/L,sodium acetate 6 g/L,ammonium citrate 5 g/L,Tween 802 mL and the initial pH 6.5,and the optimal culture conditions was the shaker speed of 120 rpm and the temperature of 30℃,the W.confusa XG-3 EPS yield was reached 61.03 g/L,an increase of 44.95%compared with before optimization.
