一株来自可口革囊星虫产纤溶酶菌株的筛选与鉴定

摘要 对野生可口革囊星虫肠道细菌进行分离,共获得10株细菌.通过琼脂-纤维蛋白平板法初筛,以透明溶圈为筛选标记,初筛出产纤溶酶菌株4株.对初筛的菌株发酵培养,培养液经琼脂-纤维蛋白平板法复筛,筛选出纤溶酶活力较高的菌株XC5.通过形态特征观察及16S rDNA序列比对分析,鉴定菌株XC5为芽胞杆菌属的Bacillus oryzaecorticis.该菌株在生长18 h后检测到最高纤溶酶活力为601.4 U·mL-1.该试验丰富了海洋来源纤溶酶菌株的菌种资源,并为其后续相关研究提供了基础. A total of 10 strains of bacteria were isolated from the intestinal bacteria of wild Phascolosoma esculenta.Four strains of fibrinolytic enzyme were screened by agar-fibrin plate method with transparent circle as screening mark.The strain XC5 with high fibrinolytic activity were screened by fermentation and culture of the initially screened strain and re-screened by agar-fibrin plate method.Through morphological characteristics observation,16S rDNA sequence alignment and phylogenetic analysis,the strain XC5 was identified as Bacillus oryzaecorticis.The highest fibrinolytic enzyme activity was 601.4 U/mL,which was detected 18 hours after its growth.This experiment enriched the strain resources of marine source plasmin strains and provided theoretical basis for its subsequent related research.
