不同麻醉剂对新生大鼠缺氧缺血性脑病模型建立的影响 被引量:4

Effects of diverse anesthetics on the establishment of the model of hypoxic ischemic encephalopathy in neonatal rats

摘要 目的探讨3种不同的麻醉剂对新生大鼠缺氧缺血性脑病(HIE)模型建立的影响。方法用Rice-Vannucci法将7日龄新生大鼠右侧颈总动脉结扎后缺氧以建立HIE模型。按照体重将大鼠随机分为6组:第1、第2和第3假手术组,第1、第2和第3模型组,每组25只。另外,按照体重将大鼠随机分为3组:第3假手术组、第3和第4模型组,每组25只。第1假手术组和第1模型组大鼠用水合氯醛进行麻醉,第2假手术组和第2模型组用巴比妥钠进行麻醉;第3假手术组和第3、第4模型组用异氟烷进行麻醉。术后,第1、第2、第3模型组缺氧1.5 h,第4模型组缺氧2 h。造模48 h后,测定各组大鼠脑含水量(BWC)和脑梗死体积(CIV)。结果第1、第2和第3假手术组的BWC%分别为(86.64±0.32)%,(87.05±0.64)%和(86.80±0.11)%;这3组的IV%均为0;第1、第2、第3和第4模型组的BWC%分别为(86.84±0.23)%,(88.25±0.57)%,(88.36±0.43)%和(89.60±0.17)%;这4组的IV%分别为(6.25±4.83)%,(15.35±4.20)%,(27.24±5.07)%和(40.62±3.50)%。上述指标:4个模型组与3个假手术组相比,IV的差异均有统计学意义(P<0.05,P<0.01);模型组的BWC较假手术组有增加趋势,但差异均无统计学意义(均P>0.05)。结论以异氟烷作为HIE造模的麻醉剂,且术后缺氧2 h的造模效果较理想。 Objective To determine the effects of three diverse anesthetics on the establishment of a model of hypoxic ischemic encephalopathy(HIE)in neonatal rats.Methods Seven days old rat pups with right common carotid artery ligation followed by hypoxia to establish HIE model by Rice-Vannucci method.Rats were randomly divided into six groups according by weight:sham-1,-2,-3 groups and model-1,-2,-3 groups.Another rats were randomly divided into three groups according by weight:sham-3 and model-3,-4 groups.The sham-1 and model-1 groups with chloral hydrate anesthesia;the sham-2 and model-2 groups with pentobarbital sodium anesthesia;sham-3 and model-3,4 groups with isoflurane anesthesia.The model-1,-2,-3 groups were exposed to hypoxia for 1.5 h and model-4 group for 2 h.Then brain water content(BWC)and cerebral infarct volume(CIV)at 48 h post HIE were measured.Results The percentage of brain water content in sham-1,-2,-3 groups were(86.64±0.32)%,(87.05±0.64)%and(86.80±0.11)%;while the percentage of cerebral infarct volume in the three groups were 0.The percentage of BWC in model-1,-2,-3,-4 groups were(86.84±0.23)%,(88.25±0.57)%,(88.36±0.43)%and(89.60±0.17)%;the percentage of cerebral infarct volume in the four groups were(6.25±4.83)%,(15.35±4.20)%,(27.24±5.07)%and(40.62±3.50)%.Comparison between four model groups and three sham groups,the difference of the CIV were significant(P<0.05,or P<0.01);BWC in model group increasing trend compared with sham operation group,but the difference of the BWC were not significant(all P>0.05).Conclusion Among the three anesthetics,isoflurane was used as the anesthetics in HIE modeling process and the model effect was better when hypoxia lasted for 2 h.
