摘要
目的研究茶多酚通过调节磷脂酰肌醇-3-激酶/蛋白激酶B(PI3-K/Akt)信号通路对甲状腺乳头状癌细胞增殖、侵袭的作用。方法取对数期人甲状腺乳头状癌K1细胞,分为低、中、高剂量实验组及对照组,分别以140,160,180 mg·L^-1茶多酚与等量生理盐水处理,分别干预24,36,48 h。用噻唑蓝法检测人甲状腺乳头状癌K1细胞增殖抑制情况;用Transwell小室实验检测人甲状腺乳头状癌K1细胞侵袭能力;用蛋白质印迹法检测人甲状腺乳头状癌K1细胞PI3-K、Akt蛋白表达。结果干预48 h时,低、中、高剂量实验组人甲状腺乳头状癌K1细胞增殖抑制率分别为(44.65±3.83)%,(52.84±3.97)%和(65.68±4.39)%;干预48 h,对照组和低、中、高剂量实验组人甲状腺乳头状癌K1细胞的侵袭率分别为(86.73±5.22)%,(67.38±4.34)%,(39.12±3.05)%,(21.62±4.23)%。随干预时间延长及茶多酚作用浓度增大,低、中、高剂量实验组人甲状腺乳头状癌K1细胞的增殖抑制率逐渐升高,且呈时间-剂量依赖性(P<0.05);与对照组比较,低、中、高剂量实验组人甲状腺乳头状癌K1细胞的侵袭率及PI3-K、Akt蛋白相对表达量均降低,且呈剂量依赖性(均P<0.05)。结论茶多酚可有效抑制人甲状腺乳头状癌K1细胞的增殖,降低其侵袭能力,其机制可能与抑制人甲状腺乳头状癌K1细胞PI3-K/Akt通路的信号转导相关。
Objective To investigate the effect of tea polyphenols on the proliferation and invasion of thyroid papillary cancer cells by regulating the phosphatidylinositol-3-kinase/protein kinase B(PI3-K/Akt)signaling pathway.Methods K1 cells of human thyroid papillary carcinoma in logarithmic phase were divided into Exp-L/M/H groups and control group,treated with 140,160,180 mg·L^-1 tea polyphenols and equivalent normal saline for 24,36,48 h.The proliferation inhibition of human thyroid papillary carcinoma K1 cells was detected by tetramethylazozol blue method;Transwell chamber test was used to detect the invasion ability of human thyroid papillary carcinoma K1 cells;Western blotting method was used to detect PI3-K and Akt protein expression in human thyroid papillary carcinoma K1 cells.Results After 48 h of intervention,the proliferation inhibition rates of human thyroid papillary carcinoma K1 cells in Exp-L/M/H groups were(44.65±3.83)%,(52.84±3.97)%,and(65.68±4.39)%.At 48 h of intervention,the invasion rates of human thyroid papillary carcinoma K1 cells in control group and Exp-L/M/H groups were(86.73±5.22)%,(67.38±4.34)%,(39.12±3.05)%,and(21.62±4.23)%.With the extension of the intervention time and the increase of the concentration of tea polyphenols,the proliferation inhibition rates of human thyroid papillary carcinoma K1 cells in Exp-L/M/H groups were gradually increased,with time-dose dependent(P<0.05).Compared with control group,the invasion rate of human thyroid papillary carcinoma K1 cells and the relative expressions of PI3-K and Akt proteins in Exp-L/M/H groups were reduced and with dose-dependent(all P<0.05).Conclusion Tea polyphenols can effectively inhibit the proliferation of human thyroid papillary carcinoma K1 cells and reduce their invasion ability,and its mechanism may be related to the inhibition of signal transduction of PI3-K/Akt pathway in human thyroid papillary carcinoma K1 cells.
