响应面法优化超声辅助溶剂提取香蕉5-羟色胺的工艺 被引量:3

Optimization of Ultrasound-Assisted Solvent Extraction of 5-Hydroxytryptamine from Banana by Response Surface Methodology

摘要 采用超声波辅助乙醇溶剂萃取法提取香蕉中的5-羟色胺,提取收率以埃尔利希试剂分光光度法测定。通过单因素试验,考察乙醇的体积分数、超声时间、超声温度以及料液比4个因素对香蕉中5-羟色胺提取收率的影响,在此基础上,利用Box-Behnken原理设计响应面法优化5-羟色胺的提取工艺条件。结果表明,5-羟色胺最佳提取工艺条件为:料液比1∶20(g/mL),乙醇体积分数72%,超声时间33 min、超声温度32℃。此工艺条件下香蕉中5-羟色胺提取收率为32.9867 mg/g,预测值和实际测定值接近。 The 5-hydroxytryptamine(5-HT)in banana was extracted by ultrasonic assisted ethanol solvent extraction,and the extraction yield was determined by Ehrlich reagent spectrophotometry.The effects of four factors including ethanol volume fraction,ultrasonic time,ultrasonic temperature and solid-liquid ratio on the extraction yield of 5-HT in banana were investigated by single factor experiment.On this basis,response surface methodology was designed to optimize the extraction process conditions of 5-HT using the Box-Behnken principle.The results showed that the optimal extraction conditions of 5-HT were as follows:1∶20(g/mL)solid-liquid ratio,ethanol volume fraction of 72%,ultrasonic time of 33 min,and ultrasonic temperature of 32℃.Under this process condition,the extraction yield of 5-HT in banana was 32.9867 mg/g,and the predicted value was close to the actual measured value.
