去甲乌药碱对过氧化氢引起的PC12细胞损伤的保护作用研究 被引量:1

摘要 目的探讨去甲乌药碱对过氧化氢(H 2O 2)引起的PC12细胞损伤的保护作用。方法将PC12细胞分为对照组、H 2O 2组(100μM H 2O 2)、去甲乌药碱组(50μM去甲乌药碱+100μM H 2O 2),CCK8法检测细胞活力,TUNEL染色检测细胞凋亡情况,流式检测ROS水平,氧化应激标志物MDA、SOD分别采用TBA法和WST法检测,蛋白免疫印迹检测凋亡相关蛋白Caspase3、Bax和Bcl-2的表达情况。结果与H 2O 2组比较,去甲乌药碱组细胞活力显著提高,ROS、MDA含量显著降低,SOD活性显著升高,Caspase3、Bax表达量减少,Bcl-2表达量增加。结论去甲乌药碱能通过抗氧化及抗凋亡抑制H 2O 2引起的PC12细胞损伤。 Objective To investigate the protective effect of higenamine on PC12 cell injury induced by hydrogen peroxide(H 2O 2).Methods PC12 cells were divided into control group,H 2O 2 group(100μM H2O2),and sulphate group(50μM Higenamine+100μM H 2O 2).Cell viability was detected by CCK8 method,apoptosis was detected by TUNEL staining,and flow detection was performed.The levels of ROS,oxidative stress markers MDA and SOD were detected by TBA and WST,respectively.Western blotting was used to detect the expression of apoptosis-related proteins Caspase3,Bax and Bcl-2.Results Compared with the H 2O 2 group,the cell viability of the sulphate group was significantly increased,the ROS and MDA contents were significantly decreased,the SOD activity was significantly increased,the expression of Caspase3 and Bax was decreased,and the expression of Bcl-2 was increased.Conclusion Higenamine can inhibit PC12 cell injury induced by H 2O 2 through anti-oxidation and anti-apoptosis.
