miR-155-5p调节Wnt信号通路促进肾透明细胞癌细胞的增殖与转移

miR-155-5p regulates Wnt signaling pathway and promotes proliferation and metastasis of renal clear cell carcinoma cells

目的研究miR-155-5p和Wnt信号通路在肾透明细胞癌细胞中的作用机制。方法采用qRT-PCR检测细胞中miR-155-5p的表达。WB检测各细胞中Wnt信号通路相关蛋白磷酸化水平及蛋白表达水平。通过MTT实验、细胞划痕实验、Transwell实验分别检测各细胞增殖、迁移和侵袭能力。结果肾透明细胞癌细胞中miR-155-5p的表达明显高于正常细胞。过表达miR-155-5p促进肾透明细胞癌细胞增殖、迁移和侵袭。加入通路蛋白抑制剂后,我们发现其会减弱过表达miR-155-5p对肾透明细胞癌细胞增殖、迁移和侵袭的促进能力。结论miR-155-5p和Wnt在调节肾透明细胞癌发生发展中起着关键作用,miR-155-5p可能成为肾透明细胞癌治疗的新靶点。

Objective Many studies have shown that miR-155-5p plays an important role in cancer.However,the role of miR-155-5p on the regulation of Wnt signaling pathway in clear cell renal cell carcinoma is less studied.The mechanism of miR-155-5p and Wnt signaling pathway in renal clear cell carcinoma cells was described.Methods qRT-PCR was used to detect miR-155-5p expression in cells.WB detected the Wnt signaling pathway-related protein phosphorylation level and protein expression level in each cell.MTT test,cell scratch test,and Transwell test were used to detect the proliferation,migration and invasion of each cell.Results In this study,we found that the expression of miR-155-5p in renal clear cell carcinoma cells was significantly higher than that in normal cells.Overexpression of miR-155-5p promotes the proliferation,migration and invasion of renal clear cell carcinoma cells.After adding a pathway protein inhibitor,we found that it weakened the ability of miR-155-5p overexpression to promote the proliferation,migration,and invasion of renal clear cell carcinoma cells.Conclusion These experiments indicate that miR-155-5p and Wnt play a key role in regulating the occurrence and development of renal clear cell carcinoma,and miR-155-5p may become a new target for the treatment of renal clear cell carcinoma.
