DOK2在早期胃癌中的表达与病理特征相关性分析

Expression of DOK2 in early gastric cancer and its correlation with pathological characteristics

目的探究酪氨酸蛋白激酶下游分子2(DOK2)在早期胃癌中的表达意义,并就DOK2表达与早期胃癌患者的病理特征相关性进行分析。方法选择2018年1月至12月于延安大学附属医院接受治疗的30例胃癌患者为实验组,采集其胃癌组织、癌旁组织,同时采集30例正常胃黏膜组织作为对照组,分析样本中DOK2mRNA表达情况,分析样本中DOK2蛋白表达情况,并对DOK2蛋白表达与胃癌临床病理学特征及其与胃癌患者生存时间相关性分析。结果①DOK2mRNA表达与DOK2蛋白表达在胃癌组织中明显低于癌旁组织和正常组织(P<0.05),癌旁组织与正常组织表达水平差异不明显(P>0.05);②DOK2蛋白表达与早期胃癌患者浸润深度、淋巴结转移和分化程度均存在密切联系(P<0.05),并与患者生存时间存在相关性(P<0.05)。结论DOK2与早期胃癌浸润状态、分化程度、淋巴结转移等病理特征存在明显相关性,可作为早期胃癌诊断及预后评估的指标之一。

Objective To explore the significance of downstream of tyrosine kinase(DOK2)expression in early gastric cancer,and to analyze the correlation between the expression of DOK2 and the pathological characteristics of patients with early gastric cancer.Methods Thirty patients with gastric cancer who were treated in our hospital from January 2018 to December 2018 were selected as the experimental group.Gastric cancer tissues and adjacent tissues were collected,and 30 normal gastric mucosa tissues were collected as the control group.The samples were analyzed.DOK2 mRNA expression,analysis of DOK2 protein expression in samples,and analysis of the correlation between DOK2 protein expression and clinicopathological characteristics of gastric cancer,and analysis of correlation between DOK2 protein expression and survival time of gastric cancer patients.Results①The expression of DOK2 mRNA in cancer tissues was significantly lower than that in adjacent tissues and normal tissues(P<0.05),and the expression levels of adjacent tissues and normal tissues were not significantly different(P>0.05).②DOK2 protein expression was closely related to the depth of invasion,lymph node metastasis and differentiation of patients with early gastric cancer(P<0.05),and correlated with survival time(P<0.05).Conclusion DOK2 has significant correlation with pathological characteristics such as invasion status,differentiation degree,and lymph node metastasis in early gastric cancer,and it can be used as one of the indicators for early gastric cancer diagnosis and prognosis evaluation.
