龟板固本方对肺癌骨髓抑制小鼠骨髓造血微环境的影响 被引量:4

Effect of Guiban Guben Recipe on Bone Marrow Hematopoietic Microenvironment in Lung Cancer Mice with Myelosuppression

摘要 目的研究龟板固本方对肺癌化疗小鼠骨髓造血微环境的影响,并探讨其作用机制。方法将60只肺癌小鼠随机分为4组,每组15只,空白组、模型组、龟板固本方组、西药组(G-CSF)。除空白组外,其他3组均建立Lewis肺癌小鼠化疗致骨髓抑制的动物模型,龟板固本方组予灌胃给药,每天0.4 ml,空白组和模型组予等量生理盐水,西药组皮下注射粒细胞集落刺激因子,各组均为连续给药7 d后,采用ELISA法及外周血象计数法等方法,检测骨髓有核细胞悬液上清中的促红细胞生成素(Erythropoietin,EPO)、血小板生成素(Thrombopoietin,TPO)和粒细胞集落刺激因子(Granulocyte Colony-stimulating Factor,G-CSF)含量和外周血常规白细胞计数(WBC)、血小板计数(PLT)和红细胞计数(RBC)。结果龟板固本方能明显升高肺癌骨髓抑制小鼠外周血的WBC、PLT和RBC计数;能提高其骨髓造血微环境的促红细胞生成素(EPO),血小板生成素(TPO),粒细胞集落刺激因子(G-CSF)含量,与模型组比较,差异均有统计学意义(P<0.01或P<0.05)。结论龟板固本方具有减轻肺癌小鼠化疗后骨髓抑制的效果,其作用机制可能与通过改善骨髓造血微环境中的造血因子EPO、TPO和G-CSF有关。 Objective The effect of Guiban Guben Recipe on bone marrow hematopoietic microenvironment in lung cancer mice with myelosuppression was observed for the exploration of its therapeutic mechanism.Methods Sixty lung cancer mice were randomly divided into 4 groups(n=15),namely normal group,model group,Guiban Guben Recipe group and western medicine G-CSF group.Except for the normal group,mice in the other three groups were induced the Lewis lung cancer model of chemotherapy-induced myelosuppression.Guiban Guben Recipe group was given intragastric administration of Guiban Guben Recipe,0.4 mL/d.The normal group and model group were given the equivalent amount of normal saline,and G-CSF group was given subcutaneous injection of granulocyte colony stimulating factor(G-CSF).After treatment for 7 continuous days,we examined the contents of erythropoietin(EPO),thrombopoietin(TPO),granulocyte colony stimulating factor(G-CSF)in the bone marrow nucleated cell suspension by ELISA and detected the peripheral blood count of white blood cell(WBC),platelet(PLT),and red blood cell(RBC)with the peripheral blood analysis.Results Guiban Guben Recipe can obviously increase the count of WBC,PLT and RBC in the peripheral blood of lung cancer mice with myelosuppression,and can increase the content of EPO,TPO and G-CSF in the hematopoietic microenvironment of bone marrow.The differences were statistically significant in comparison with the model group(P<0.01 or P<0.05).Conclusion Guiban Guben Recipe exerts certain effect on reducing the chemotherapy-induced myelosup pression in lung cancer mice,and its mechanism may be related to the improvement of hematopoietic factors ofEPO,TPO and G-CSF in hematopoietic microenvironment of bone marrow.
