高效液相色谱法测定特殊医学用途婴幼儿配方食品中乳铁蛋白含量 被引量:4

摘要 本研究以食品安全国家标准《食品中乳铁蛋白的测定(征求意见稿)》为参考,优化预处理步骤和梯度洗脱条件,建立了一种用紫外高效液相色谱测定特殊用途婴幼儿配方食品中乳铁蛋白含量的方法。色谱柱为C4,5μm,300A,250 mm×4.6 mm,流动相A为乙腈,流动相B为0.1%三氟乙酸,检测波长为280 nm。结果表明,乳铁蛋白的检出限为2.3 mg/100 g,定量限为3.9 mg/100 g,在乳铁蛋白浓度为10~140μg·mL^-1范围内,检测结果呈良好线性关系,回收率在98%~105%。用此方法检测婴幼儿配方奶粉中乳铁蛋白含量,操作简单,结果准确可靠,精密度高,重现性好。 This study was based on the national food safety standards food on the determination of lactoferrin,optimizing the pretreatment steps and gradient elution conditions,establishing a method with a HPLC to determin the lactoferrin containingin infant special medical purposes formula food Chromatographic conditions:the chromatographic column was C4,5μm,300A,250 mm×4.6 mm,the mobile phase A was acetonitrile,the mobile phase B was 0.1%trifluoroacetic acid,and the detection wavelength was 280 nm.The results showed that the detection limit of lactoferrin was 2.5 mg/100 g,and the determination limit was 4.0 mg/100 g which is in the range of lactoferrin concentration of 10~140μg·mL^-1.The detection results showed a good linear relationship,and the recovery rate was 98%~105%.The method on determining lactoferrin content in infant formula has more convenient operation,higher precision,higher accuracy and better repeatability.
