奶粉摄入改善慢性不可预知性应激诱导的小鼠抑郁样行为

摘要 探讨奶粉摄入对慢性不可预知性应激诱导的小鼠抑郁样行为的影响。选取雄性ICR小鼠40只,随机分为对照组、抑郁模型组、抗抑郁药物组和奶粉组。对后三组小鼠施加慢性不可预知性应激6周,第4周验证抑郁模型,随后2周施加盐酸氟西汀或奶粉干预。监测小鼠体重变化与摄食情况,HE染色观察小鼠海马CA1区形态学变化,Western Blot检测小鼠海马区过氧化物酶增殖物激活受体(Peroxisome proliferator-activated receptorα,PPARα)与血红素氧合酶(heme oxygenase-1,HO-1)蛋白表达水平。发现慢性不可预知性应激诱导小鼠抑郁样行为,降低小鼠体重(P<0.5),改变大脑海马体形态学结构,给予抗抑郁药物或奶粉后,体重回升且小鼠抑郁样行为有所改善(P<0.5),小鼠海马区PPARα与血HO-1蛋白表达水平被逆转(P<0.5)。认为奶粉摄入可改善慢性不可预知性应激诱导的小鼠抑郁样行为,可能与PPARα与HO-1的表达有关。 To explore the effect of milk powder on depression-like behavior in mice induced by chronic unpredictable stress.Forty male ICR mice were randomly devided into control group,depression model group,antidepression drug group and milk powder group.Chronic unpredictable stress was applied to the later three groups of mice for 6 weeks,and the depression model was validated at week 4,followed by fluoxetine hydrochloride or milk powder intervention for 2 weeks.The weight and feeding of mice were monitored.HE staining was used to observe the morphological changes of CA1 area in hippocampus of mice.Protein levels of peroxisome proliferator-activated receptorα(PPARα)and heme oxygenase-1(HO-1)were detected in hippocampus of mice by Western Blot.Result Chronic unpredictable stress induced depression-like behavior in mice,reduced body weight(P<0.5)and changed the morphology of the hippocampus.Body weight increased after administration of antidepressants or milk powder,the depression-like behavior of mice was improved(P<0.5),and protein expressions of PPARαand HO-1 in hippocampus of mice were reversed(P<0.5).Conclusion Milk powder can improve depression-like behavior in mice induced by chronic unpredictable stress,which may be closely related to the protein expression of PPARαand HO-1.
