超高效液相色谱-串联质谱法测定运动型乳饮品中的氨基酸 被引量:3

摘要 建立了一种超高效液相色谱-串联质谱法测定运动型乳饮品中16种氨基酸的方法。样品提取液经固相萃取小柱进行净化和富集,过滤膜后上机检测。结果表明,以甲醇-0.1%甲酸溶液为流动相,在梯度洗脱模式下,16种氨基酸在0.10~10.00μmol/L浓度范围内线性良好,相关系数r2为0.9924~0.9993,检出限范围为0.005~0.05μmol/L,定量限范围为0.03~0.20μmol/L,加标回收率为85.1%~96.2%,相对标准偏差(RSD)为1.1%~5.2%。该方法具有前处理简单、灵敏度高和检测速度快的优点,适用于运动型乳饮品中16种氨基酸的定量检测。 A method for the determination of 16 amino acids in sports milk drinks by ultra-high performance liquid chromatography-tandem mass spectrometry was established.Sample extract was purified and enriched by solid phase extraction column,and then detected on the machine after filtration membrane.The results showed that 16 amino acids had good linearity in the concentration range of 0.10~10.00μmol/L under gradient elution mode with methanol-0.1%formic acid solution as mobile phase.The correlation coefficient r2 was 0.9924~0.9993,the detection limit was 0.005~0.05μmol/L,the quantitative limit was 0.03~0.20μmol/L,and the recovery was 85.1%~96.2%,and the relative standard deviation(RSD)was 1.1%~5.2%.The method has the advantages of simple pretreatment,good purifying effect,high sensitivity and fast detection rate,and is suitable for the quantitative determination of 16 amino acids in sports milk drinks.
