酸柏栀油软胶囊对学习记忆障碍模型小鼠行为学及自由基代谢的影响 被引量:1

Effects of Suanbaizhiyou Soft Capsule on Ethology and Free Radical Metabolism in Mice with Learning and Memory Disorder

摘要 目的:观察酸柏栀油软胶囊对学习记忆障碍模型小鼠的行为学与自由基代谢的影响。方法:选购昆明种小鼠96只,适应性喂养1周后,随机分为空白对照组、模型组、酸柏栀油软胶囊高剂量组(5 mL·kg^-1)、酸柏栀油软胶囊中剂量组(2.5 mL·kg^-1)、酸柏栀油软胶囊低剂量组(1.25 mL·kg^-1)、阴性对照组、吡拉西坦组、DHA组,每组12只。灌胃1 h后,空白组给予腹腔注射生理盐水4 mL·kg^-1,其他组制备小鼠记忆获得障碍模型,然后对应药物灌胃,每日1次,连续40 d。用药结束后经Morris水迷宫实验对小鼠行为学进行检测,酶联免疫吸附法检测小鼠脑组织中自由基代谢水平。结果:第1-5天,第6天,与其他组比较,酸柏栀油软胶囊高剂量组小鼠寻找平台潜伏期及寻找平台路径最短(P<0.05);且随着酸柏栀油软胶囊剂量增加,小鼠寻找平台潜伏期及寻找平台路径缩短。与其他组比较,酸柏栀油软胶囊高剂量组小鼠第Ⅲ象限停留时间最短、第Ⅲ象限路程百分比最低、穿台次数最多(P<0.05),且随着酸柏栀油软胶囊剂量增加,小鼠第Ⅲ象限停留时间缩短、第Ⅲ象限路程百分比降低、穿台次数升高(P<0.05)。酸柏栀油软胶囊高剂量组小鼠血清、脑组织中超氧化物歧化酶、总抗氧化能力、一氧化氮、丙二醛水平显著高于其他组(P<0.05),且随着酸柏栀油软胶囊剂量增加,超氧化物歧化酶、总抗氧化能力、一氧化氮、丙二醛水平升高(P<0.05)。结论:酸柏栀油软胶囊可通过提高机体清除自由基的能力,降低脂质过氧化损伤程度,保护脑组织,达到改善模型小鼠学习记忆能力的目的。 Objective:To observe the effect of Suanbaizhiyou soft capsule on ethology and free radical metabolism in mice with learning and memory disorder.Methods:96 Kunming mice were purchased and fed for 1 week.They were randomly divided into blank control group,model group,Suanbaizhiyou soft capsule high-dose group(5 mL·kg^-1),Suanbaizhiyou soft capsule medium-dose group(2.5 mL·kg^-1),Suanbaizhiyou soft capsule low-dose group(1.25 mL·kg^-1),negative control group,piracetam group,DHA group,12 mice in each group.After 1 hour of intragastric administration,the blank group was given intraperitoneal injection of 4 mL·kg^-1 of normal saline.The other groups prepared mice with memory disorder models,and then the corresponding drugs were given intragastrically once a day for 40 consecutive days.After the treatment,the ethology of mice was tested by Morris water maze experiment,and the level of free radical metabolism in the brain tissue of mice was tested by enzyme-linked immunosorbent assay.Results:On the first to fifth day and the sixth day,compared with other groups,mice in the Suanbaizhiyou soft capsule group had the shortest platform latency and the shortest path for finding a platform(P<0.05);and with the increase in the mass concentration of Suanbaizhiyou soft capsule,the incubation period and the path of finding a platform for mice are shortened.Compared with other groups,mice in the Suanbai Zhiyou soft capsule group had the shortest residence time in the III quadrant,the lowest percentage of distance in the III quadrant,and the most number of trips(P<0.05),and with the increase in the mass concentration of the Suanbai Zhiyou soft capsule,the mouse stays in the III quadrant shortened,the distance percentage in the III quadrant decreased,and the number of crossings increased(P<0.05).The level of mice serum,brain tissue superoxide dismutase(SOD),total antioxidant capacity(T-AOC),nitric oxide(NO)and malondialdehyde(MDA)in Suanbaizhiyou soft capsule group was significantly higher than those of the other groups(P<0.05),and with the increase in the mass concentration of Suanbaizhiyou soft capsule,the levels of SOD,T-AOC,NO and MDA increased(P<0.05).Conclusion:Suanbaizhiyou soft capsule can improve the body′s ability to scavenge free radicals,reduce lipid peroxidation damage,protect brain tissue,and achieve the purpose of improving the learning and memory ability of model mice.
