摘要
目的:研究四藤方对MGC-803胃癌生长的影响,并探讨可能的作用机制。方法:体外培养人胃癌细胞MGC-803,建立人胃癌MGC-803裸小鼠皮下移植瘤模型。随机分组,分为对照组、西药组和中药组,每组10只;对照组生理盐水灌胃,每次0.3 mL,每天1次;西药组5-氟尿嘧啶(5-fluorouracil,5-FU)腹腔注射,每次1 mg,每周1次;中药组采用四藤方煎剂灌胃,每次0.3 mL,每天1次,含生药1 g·mL^-1。共治疗8周。治疗期间记录肿瘤生长变化。治疗结束后脱颈椎处死实验动物,剥离肿瘤称量。免疫组织化学法检测Smac/Survivin信号蛋白表达水平。结果:8周治疗后,5-FU和四藤方组裸小鼠皮下移植瘤体积、瘤质量均小于对照组,且移植瘤体积增长也较对照组体积增长缓慢。5-FU和四藤方均能明显上调Caspase-3和Caspase-7蛋白表达,下调Survivin蛋白水平,升高Smac蛋白表达。结论:四藤方可抑制MGC-803胃癌细胞生长,其机制可能与通过调控Smac/Survivin信号转导通路蛋白表达相关。
Objective:To study the effect of Si Teng Prescription on the growth of MGC-803 gastric cancer;and to explore the possible mechanism of action.Methods:Human gastric cancer cell MGC-803 was cultured in vitro,and a subcutaneous transplanted tumor model of human gastric cancer MGC-803 was established in nude mice.Randomized groups were divided into control group,western medicine group and traditional Chinese medicine group,each with 10 rats;the control group was intragastrically administered with normal saline,0.3mL each time,once a day;the western medicine group 5-fluorouracil(5-FU).Intraperitoneal injection was carried out,1 mg each time,once a week;the traditional Chinese medicine group was given Si Teng Prescription by gavage,0.3 mL each time,once a day,containing crude drug 1 g·mL^-1;treatment for 8 weeks in total.Observe the changes in tumor growth during treatment.After the treatment,the experimental animals were killed by removing the neck cone,and the tumor was stripped and weighed.The immunohistochemical method was used to detect the expression level of Smac/Survivin signal protein.Results:After 8 weeks of treatment,the volume and mass of subcutaneous transplanted tumors in nude mice in 5-FU and Si Teng Prescription groups were smaller than those in the control group,and the growth of transplanted tumor volume was also slower than that in the control group.Both 5-FU and Si Teng Prescription can significantly up-regulate the expression of Caspase-3 and Caspase-7 protein,down-regulate the level of Survivin protein,and increase the expression of Smac protein.Conclusion:Si Teng Prescription can inhibit the growth of MGC-803 human gastric cancer,and its mechanism may be related to the regulation of Smac/Survivin signal transduction pathway protein expression.
