摘要
建立HPLC测定方法,并结合化学计量学对5种产地掌叶大黄中蒽醌类成分进行比较分析。采用XBridge C 18(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.1%磷酸溶液为流动相,梯度洗脱,流速1.0 mL/min,检测波长254 nm,柱温30℃;芦荟大黄素、大黄酸、大黄素、大黄酚、大黄素甲醚进样量线性范围分别为43.7~1398.4 ng(r=0.9999)、41.65~1332.8 ng(r=0.9999)、40.35~1291.2 ng(r=0.9999)、40.9~1308.8 ng(r=0.9999)、21.7~694.4 ng(r=0.9998);精密度、稳定性、重复性实验的RSD<2%;该方法操作简便,精密度、稳定性、重复性好,可用于大黄药材中蒽醌类成分的含量测定;不同产地的掌叶大黄药材质量差异较大。
An HPLC method for the determination of anthraquinone components in Rheum palmatum from 5 habitats combined with chemometrics methods was developed.The HPLC method was performed on a XBridge C 18 column(4.6 mm×250 mm,5μm)with the gradient elution of methanol-0.1%phosphoric acid at the flow rate of 1.0 mL/min.The detection wavelength was set at 254 nm and the column temperature was 30℃.Aloe-Emodin,rhein,emodin,chrysophanol and physcion revealed a good linear relationship in the range of 43.7~1398.4 ng(r=0.9999),41.65~1332.8 ng(r=0.9999),40.35~1291.2 ng(r=0.9999),40.9~1308.8 ng(r=0.9999)and 21.7~694.4 ng(r=0.9998),respectively.RSDs of precision,stability and reproducibility tests were lower than 2%.The method is simple,precise,stable and reproducible,and it can be used for the anthraquinone components in medicinal material.The quality of Rheum palmatum from different regions were quite different.
