采用他克莫司建立免疫抑制小鼠模型

Application of Tacrolimus for the Establishment of Immunosuppressive Mice Model

目的探讨采用他克莫司建立免疫抑制小鼠模型的方法和效果,为实验研究提供合适的免疫抑制动物模型。方法小鼠腹腔注射他克莫司,注射剂量为1 mg·kg^-1·d^-1,连续注射2周,通过检测胸腺和脾脏指数、外周血CD4^+/CD8^+T细胞亚群比例、脾脏T细胞增殖能力,进行免疫抑制模型的鉴定。采用两独立样本t检验进行统计分析。结果模型组小鼠出现反应迟钝、毛色晦暗等一般症状。与对照组相比,模型组小鼠胸腺和脾脏指数降低(P<0.05);模型组小鼠外周血CD4^+/CD8^+T细胞亚群比例为3.24±0.28,低于对照组小鼠(1.67±0.39)(P<0.05);模型组小鼠脾脏T细胞的增殖指数低于对照组(P<0.05)。结论他克莫司可以成功建立免疫抑制的小鼠模型。

Objective To explore the method and outcome of establishing an immunosuppressive mouse model by tacrolimus,and to provide suitable immunosuppressive animal model for experimental study.Methods Mice were intraperitoneally injected with tacrolimus(1 mg·kg^-1·d^-1)for two weeks.Comprehensive evaluation was performed by thymus index and spleen index.The percentage of T lymphocyte subtype in peripheral blood and the proliferation of spleen T lymphocyte were also examined to evaluate the immune status of animal mice.The two independent samples t test was performed for statistical analysis.Results The mice in the model group presented general symptoms such as retarded response and dark hair.Compared with the control group,the thymus and spleen index of mice in the model group decreased(P<0.05).The proportion of peripheral blood CD4^+/CD8^+T cell subsets in model group was 3.24±0.28,lower than that in control group(1.67±0.39)(P<0.05).The proliferation of spleen T cell was significantly decreased in tacrolimus group than that in normal group(P<0.05).Conclusion Application of tacrolimus can successfully establish immunosuppressive mice model.
