摘要
目的:通过建立小鼠模型,探讨蛔虫感染在过敏性哮喘小鼠中的免疫机制。方法:60只雄性SPF小鼠随机分成蛔虫感染组、过敏性哮喘组、蛔虫感染合并过敏性哮喘组和正常对照组,每组15只。采用直接计数法计数外周血嗜酸性粒细胞(EOS);免疫比浊法测定血清总IgE;ELISA测定各组小鼠血清中IL-4、IL-10、TNF-α、INF-Υ的水平;流式细胞仪检测外周血中CD3、CD4、CD8淋巴细胞绝对数及CD4/CD8比值。结果:蛔虫感染组、过敏性哮喘组和蛔虫感染合并过敏性哮喘组的嗜酸性粒细胞计数(EOS)、血清总IgE、IL-4、IL-10、TNF-α,CD3、CD4与正常对照组比较差异均有统计学意义(P<0.05)。结论:蛔虫感染可造成宿主明显的嗜酸性粒细胞数升高、血清总IgE水平升高、细胞因子IL-4、IL-10的表达以及CD3、CD4细胞的升高,但在合并过敏性哮喘时可能对宿主免疫系统起抑制作用,可降低哮喘的发生。
Objective:To explore the immune mechanism of Ascaris infection in allergic asthmatic mice by establishing mice model.Methods:60 male SPF mice were randomLy divided into three groups:Ascaris infection group,allergic asthma group,Ascaris infection with allergic asthma group and normal control group.Eosinophils in peripheral blood were counted by direct counting method;total IgE in serum was measured by immunoturbidimetry;the levels of IL-4,IL-10,TNF-α,INF-γin serum were measured by ELISA;the absolute numbers of CD3,CD4,CD8 lymphocytes and the ratio of CD4/CD8 in peripheral blood were measured by flow cytometry.Results:Compared with the control group,EOS,total IgE,IL-4,IL-10,TNF-α,CD3 and CD4 in Ascaris infection group and allergic asthma group were statistically significant(P<0.05).Conclusion:Ascaris infection can increase the number of eosinophils,the level of serum total IgE,the expression of cytokines IL-4 and IL-10,and the increase of CD3 and CD4 cells,but it may inhibit the host immune system and reduce the incidence of asthma in combination with allergic asthma.
