化瘀消癥软胶囊对子宫肌瘤大鼠的干预作用及其机制 被引量:2

摘要 目的探究化瘀消癥软胶囊对子宫肌瘤大鼠的干预作用及其机制。方法将60只雌性SD大鼠随机分为空白组、模型对照组、化瘀消癥软胶囊低、中、高剂量组和阳性对照米非司酮组,每组10只;除空白组外,其余各组均采用雌激素负荷法构建子宫肌瘤大鼠模型。药物干预后,测定大鼠子宫系数,ELISA法检测大鼠血清雌二醇(E2)、孕酮(P)、黄体生成素(LH)和卵泡刺激素(FSH)水平,HE染色观察大鼠子宫平滑肌组织病理学变化,Western blot检测大鼠子宫组织碱性成纤维细胞生长因子(bFGF)、缺氧诱导因子-1α(HIF-1α)和血管内皮生长因子(VEGF)等蛋白表达水平。结果与空白组比较,模型对照组大鼠子宫平滑肌增生明显,子宫系数显著增高(P<0.05),血清中E2、P水平显著上升(P<0.05),而LH、FSH水平显著降低(P<0.05),且子宫组织中bFGF、HIF-1α和VEGF等蛋白表达水平显著增加(P<0.05)。与模型对照组比较,化瘀消癥软胶囊高剂量组和米非司酮组大鼠子宫平滑肌增生被抑制,子宫系数显著降低(P<0.05),血清中E2、P水平显著降低(P<0.05),而LH、FSH水平显著增加(P<0.05),且子宫组织中bFGF、HIF-1α和VEGF等蛋白表达水平显著降低(P<0.05)。结论化瘀消癥软胶囊对子宫肌瘤有明显的抑制作用,其作用机制可能与改善子宫病理学变化、调节性激素水平、抑制血管生成等有关。 Objective:To explore intervention effect of Huayu Xiaozheng soft capsule(HXSC)on hysteromyoma in rats and its mechanism.Methods:Sixty female SD rats were randomly divided into control group,model group,low-dose,medium-dose and high-dose HXSC group,and positive control mifepristone group,with 10 rats in each group.Except control group,estrogen load method was applied in the remaining groups to establish model of rat with hysteromyoma.After drug intervention,the uterine coefficient of rats was detected.The serum levels of estradiol(E2),progesterone(P),LH and FSH in rats were detected by ELISA method.The histopathological changes of uterine smooth muscle of rats were observed by HE staining.The protein expression levels of basic fibroblast growth factor(bFGF),hypoxia-inducible factor-1(HIF-1α)and vascular endothelial growth factor(VEGF)in uterine tissue of rat were detected by Western blot.Results:Compared with the control group,the uterine smooth muscle of the model group was significantly proliferated and the uterine coefficient was significantly increased(P<0.05);the serum levels of E2 and P were significantly increased(P<0.05),while the serum levels of LH and FSH were significantly reduced(P<0.05);the protein expression levels of bFGF,HIF-1αand VEGF in uterine tissue were significantly increased(P<0.05).Compared with model group,uterine smooth muscle hyperplasia in high-dose HXSC group and mifepristone group was inhibited,and the uterine coefficient was significantly reduced(P<0.05),the serum levels of E2 and P were significantly reduced(P<0.05),while the levels of LH and FSH were significantly increased(P<0.05),and the protein expression levels of bFGF,HIF-1αand VEGF in the uterine tissue were significantly reduced(P<0.05).Conclusions:HXSC has significant inhibitory effect on hysteromyoma,and its mechanism may be related to improvement of uterine pathological changes,regulation of sex hormone levels,and inhibition of angiogenesis.
