刺果番荔枝叶多酚提取工艺优化及其体外抗氧化活性 被引量:12

Optimization of Extraction Process of Polyphenols fromAnnona squamosa Leaves and Its Antioxidant Activity in Vitro

摘要 本试验选用原料为刺果番荔枝叶,使用乙醇溶剂提取其中的多酚类物质,分别比较乙醇体积分数、提取温度、液料比、提取时间4个要素对多酚提取量的影响,并采用响应面法中的Box-Benhnken中心组合试验确定最优提取工艺。采用DPPH自由基、羟自由基(·OH)法研究刺果番荔枝叶多酚抗氧化活性,比较在最优工艺条件下提取的刺果番荔枝叶多酚与抗坏血酸的清除能力。结果表明:乙醇体积分数55%、液料比50∶1(mL/g)、提取时间96 min、提取温度60℃时,多酚提取量最大,达到(20.37±0.34)mg/g。刺果番荔枝叶多酚对DPPH自由基、羟自由基(·OH)的半抑制浓度(IC50)分别为133.33、264.65μg/mL,样品在试验范围内的最大清除率分别为(51.34%±2.68%)、(52.50%±2.29%)。综上表明,刺果番荔枝叶多酚具有较好的抗氧化能力。 In this experiment,the raw material was Annona squamosa leaves,and the polyphenol substances in the leaves were extracted by using ethanol solvent.The effects of ethanol volume fraction,extraction temperature,liquid-solid ratio and extraction time on the polyphenol extraction amount were respectively compared.The Box-Benhnken center combination test in response surface methodology was used to determine the optimal extraction process.DPPH radical and hydroxyl radical(·OH)methods were used to study the antioxidant activity of polyphenols from Annona squamosa leaves,and the scavenging ability of polyphenols from Annona squamosa leaves and ascorbic acid extracted under the optimal process conditions were compared.The results showed that when the volume fraction of ethanol was 55%,the ratio of liquid to solid was 50∶1(mL/g),the extraction time was 96 min,and the extraction temperature was 60℃,the extraction amount of polyphenol was the largest,reached(20.37±0.34)mg/g.The semi-inhibitory concentrations(IC50)of polyphenols from Annona squamosa leaves on DPPH radical and hydroxyl radical(·OH)were 133.33 and 264.65μg/mL respectively.The maximum scavenging rates of the samples in the test range were(51.34%±2.68%)and(52.50%±2.29%)respectively.In summary,the polyphenols from Annona squamosa had good antioxidant capacity and would provide theoretical basis for the development of polyphenols from Annona squamosa.
