水貂皮肤黑色素细胞体外分离、培养及初步鉴定 被引量:1

摘要 为了探究黑色素细胞在水貂毛色形成过程中的生物学功能,为毛色基因功能的深入研究提供适宜的细胞模型。本研究采用0.25%DispaseⅡ、0.1%胶原酶Ⅰ及0.25%胰蛋白酶(含0.02%EDTA)联合消化法获取水貂皮肤黑色素细胞,利用不同培养基进行选择培养、传代培养与纯化,并采用多巴染色法染色,进一步对其开展了形态学初步鉴定。结果表明,酶消化分离的水貂黑色素细胞在M-254培养基中生长良好,显微镜观察细胞呈两极或三极状,多巴染色鉴定黑色素细胞呈棕黑色阳性,初步鉴定为水貂黑色素细胞。本研究为深入揭示水貂皮肤黑色素细胞在毛色形成过程中的作用奠定了理论基础。 In order to explore the biological functions of melanocytes in the process of mink hair color formation,and provide a suitable cell model for the in-depth study of hair color gene functions,the melanocytes of mink skin were isolated.In this study,the combined digestion method of 0.25%DispaseⅡ,0.1%collagenaseⅠand 0.25%pancreatin(containing 0.02%EDTA)was used to obtain melanocytes from mink skin.Different media were used for selective culture,subculture and purification.Further preliminarymorphological identification was carried out by DOPA staining.The results showed that the melanin cells isolated by enzyme digestion grew well in M-254 medium,which were bipolar or tripolar under the microscopic observation,and brown black positive in DOPAstaining.The cells were identified as mink melanocytes.This study provided more information for further revealing the role of mink melanocytes in the process of hair color formation.
