犬Ⅰ型腺病毒E1A蛋白的生物信息学分析 被引量:1

摘要 为进一步了解犬Ⅰ型腺病毒,并深入探究E1A蛋白的基本结构,为CAdV-Ⅰ的研究奠定基础。该研究利用Protparam、ProtScale、SignalP 5.0 Server和TMHMMServer 2.0等在线服务器对E1A蛋白的理化性质、信号肽和二级及三级结构等进行预测。结果发现,E1A蛋白氨基酸数量为225个;等电点为11.86,带正电荷氨基酸残基数和带负电荷氨基酸残基数分别为36和2;E1A蛋白为有跨膜结构、疏水性蛋白、无信号肽、无糖基化位点、含有9个丝氨酸磷酸化位点,10个苏氨酸磷酸化位点,不存在酪氨酸磷酸化位点;-螺旋和无规则卷曲结构是E1A蛋白的二级结构中最主要的元件,可信度为19.0%,蛋白覆盖率为9%,有6个抗原决定簇。以上结果有利于进一步开展E1A蛋白的研究,为建立CAdV-Ⅰ的抗体检测方法和亚单位疫苗提供了基础。该研究运用生物信息学分析方法预测E1A蛋白,但生物信息学是在已有知识基础上的预测,所以未来还需要对此进行进一步研究探讨。 This experiment was conducted to further understand the canineⅠadenovirus,explore the basic structure of E1A protein,and lay the foundation for the research of CAdV-Ⅰ.The online servers such as Protparam,ProtScale,SignalP5.0Server and TMHMMServer 2.0 were used to predict the physical and chemical properties,signaling peptides and secondary and tertiary structures of E1Aprotein.The amino acid number of E1A protein is 225,the isoelectric point is 11.86,the positively-charged amino acid base of residual base and negativelycharged amino acids are 36 and 2,respectively.E1Aprotein is hydrophobic protein with a membrane-across structure,without signal peptide and glycosylation sites,and it contains nine serine phosphorylation sites and 10 threonine phosphorylation sites.But there is no tyrosine phosphorylation site.The alpha helix and random curly structure are the main components of E1A protein's secondary structure,the credibility is 19.0%,the protein coverage rate is 9%.There are six antigenic determinants.These results are helpful for further research on E1A proteins,and provide the foundation for establishing the antibody detection method and subunit vaccine of CAdV-Ⅰ.In this experiment,the bioinformatics analysis method was used to predict E1A protein.However,bioinformatics is a prediction based on the existing knowledge,so further research and discussion on this topic are needed in the future.
