菲律宾蛤仔共附生细菌Pseudoalteromonas sp.JP多糖絮凝剂的分离纯化及表征

摘要 选取从菲律宾蛤仔黏附污泥中分离出的絮凝活性菌株Pseudoalteromonas sp.JP,从该菌株发酵液中提取活性多糖,使用deae-52离子交换柱和Sephadex分子筛分离得到多糖纯品,对其进行红外光谱分析,发现其含有多糖类物质的典型特征,主要基团包括羰基、羟基、碳骨架基团等,其中羰基、羟基的存在可能对絮凝分子的吸附效果有促进作用。另外,对胞外多糖纯品的絮凝活性和脱色活性进行了测定,结果显示,该微生物胞外多糖絮凝剂用量较少,且有着较好的脱色效果,在剂量为在0.8 mg·L^-1时,最佳絮凝效果达到83.5%,在反应剂量为1.5 mg·L^-1时,对孔雀石绿的最佳脱色效果为93.6%。 In this paper,the flocculating active strain Pseudoalteromonas sp.JP isolated from the associated mud of philippinarum was selected.The active polysaccharide was extracted from the fermentation broth of the strain,and the pure polysaccharide was separated by deae-52 ion exchange column and Sephadex column.The infrared spectrum was analyzed and found to contain typical characteristics of polysaccharides.The main groups include carbonyl,hydroxyl group,carbon skeleton group and so on.The presence of carbonyl and hydroxyl groups may promote the adsorption effect of flocculating molecules.In addition,the flocculating activity and decolorization activity of the pure extracellular polysaccharides were measured.The results showed that the microbial exopolysaccharide flocculant was used in a small amount and had a good decolorizing effect.When the dosage was 0.8 mg·L^-1,The best flocculation effect was 83.5%.At the reaction dose of 1.5 mg·L^-1,the best decolorization effect on malachite green was 93.6%.
