摘要
目的探究脊柱结核潜在的生物标记物,为脊柱结核的大范围筛查、早期诊断提供辅助依据。方法运用生物学技术和信息学分析,研究了青海地区(结核高发地区)脊柱结核患者不同民族(藏族、回族和汉族)间TNF-α308、IFNG rs2430561、IL-10(-1082G/A,-592C/A)、IL-12(IL12RB1 rs436857)位点的基因多态性。结果青海地区藏族脊柱结核患者中TNF-α308 A、IFNG rs2430561 A、IL-10-1082 A、IL12RB1 rs436857 A等位基因均存在差异性表达(过度或抑制),其基因多态性与青海地区藏族脊柱结核的发病可能相关;IL-10-592C/A基因多态性与青海地区藏族脊柱结核的发病可能没有关系。TNF-α308、IFNG rs2430561、IL-10(-1082G/A,-592C/A)、IL12RB1 rs436857基因多态性与青海地区藏、回族脊柱结核的发病可能无相关性。结论TNF-α308、IFNG rs2430561、IL-10(-1082G/A,-592C/A)、IL12RB1 rs436857基因多态性与脊柱结核发病可能与病人所属民族有关。
Objective To explore the potential biomarkers of spinal tuberculosis and provide a basis for the extensive screening and early diagnosis of spinal tuberculosis. Methods We studied the gene polymorphisms at the loci of TNF-"308,IFNG rs2430561,IL-10(-1082 G/A,-592 C/A) and IL-12( IL12 RB1 rs436857) among different nationalities( Tibetans,Huis and Han) of spinal tuberculosis patients in Qinghai( high incidence area of tuberculosis) by using biological technology and informatics analysis. Results TNF-" 308 A,IFNG rs2430561 A,IL-10-1082 A,IL12 RB1 rs436857 A alleles were differentially expressed( overexpression or inhibition) in Tibetan patients with spinal tuberculosis in Qinghai area,and the polymorphism of these genes may be related to the occurrence of spinal tuberculosis in Qinghai area. The incidence of spinal tuberculosis in Tibetans may not be related with IL-10-592 C/A gene polymorphism. TNF-α308,IFNG rs2430561,IL-10(-1082 G/A,-592 C/A),IL12 RB1 rs436857 gene polymorphisms may not be associated with the incidence of spinal tuberculosis in Tibetan and Hui nationalities in Qinghai. Conclusion TNF-α308,IFNG rs2430561,IL-10(-1082 G/A,-592 C/A),IL12 RB1 rs436857 gene polymorphisms and the incidence of spinal tuberculosis may be related to the nationality of the patients.
