摘要
泛素-蛋白酶途径可调节多种细胞过程,近年来以该途径为导向的研究逐渐增多。特异性蛋白酶7作为其调节泛素化动力学的关键成分——去泛素化酶家族成员之一,参与病毒复制、肿瘤、神经系统病变、炎症反应等多种疾病的相关蛋白调控。在过去十多年进行了一系列的相关疾病及其抑制剂研究,一些抑制剂已经初步显现出其对肿瘤、炎症等疾病的治疗意义。本文就特异性蛋白酶7的结构、功能及其与疾病的关联和其抑制剂进行综述。
The ubiquitin-protease pathway is involved in the regulation of various cellular processes as a post-translational modification of proteins.Specific protease 7(USP7)is a member of the deubiquitinase family that regulates ubiquitination kinetics.Currently,it has been found that USP7 can participate in the regulation of proteins related to various diseases such as viral replication,tumor proliferation,neuropathy,and inflammatory response.In the past decade,a series of researches on related diseases and inhibitors have been carried out.Some of the inhibitors have preliminarily shown their therapeutic significance for tumor,inflammation and other diseases.This paper summarizes the research progress of the structure,function of USP7,related diseases and inhibitors.
