新生猪缺氧缺血脑损伤后神经网络重建相关蛋白的表达

摘要 目的检测新生猪缺氧缺血(HI)脑损伤模型在损伤后不同时间点突触素(Syn)和神经蛋白聚糖(Neu)含量的变化,以期进一步阐明HI损伤后神经网络重建的机制。方法将28头3~5日龄的新生猪随机为对照组(n=4)和HI模型组(n=24)。HI模型组根据HI后的存活时间分为6个亚组,分别是0~2 h亚组、2~6 h亚组、6~12 h亚组、12~24 h亚组、24~48 h亚组,48~72 h亚组,每个亚组4头。通过免疫组化染色检测对照组和HI模型组各亚组Syn和Neu含量的变化。结果HI后Syn的表达整体呈先升高后降低的趋势,6~12 h表达达到高峰,而后下降。与对照组相比,HI模型组0~2 h亚组、2~6 h亚组、6~12 h亚组Syn的表达均升高,差异均有统计学意义(P<0.05);与HI模型组6~12 h亚组相比,HI模型组12~24 h亚组、24~48 h亚组、48~72 h亚组Syn的表达均降低,差异均有统计学意义(P<0.05)。HI后Neu的表达整体呈先降低后升高再降低的趋势,6~12 h表达最低,而后升高,24~48 h表达达到高峰,随后下降。与HI模型组6~12 h亚组相比,对照组及HI模型组0~2 h亚组、2~6 h亚组、12~24 h亚组、24~48 h亚组、48~72 h亚组Neu的表达均升高,差异均有统计学意义(P<0.05);与HI模型组24~48 h亚组相比,对照组及HI模型组0~2 h亚组、2~6 h亚组、6~12 h亚组、12~24 h亚组、48~72 h亚组Neu的表达均降低,差异均有统计学意义(P<0.05)。结论HI损伤可影响脑组织Syn和Neu的表达,损伤后脑神经具有较强的自我修复能力。 Objective To clarify the mechanism of neural network reconstruction after hypoxic-ischemic(HI)brain injury by detecting the changes in synaptophysin(Syn)and neurocan(Neu)levels in neonatal piglets at different time points after HI injury.Methods Twenty-eight neonatal piglets(3-5 days)were randomly divided into control(n=4)and HI model(n=24)groups.Further,the HI model group was divided into six subgroups according to the survival time after HI(0-2,2-6,6-12,12-24,24-48,and 48-72 h,n=4 each subgroup).Immunohistochemical staining was performed to detect the changes in Syn and Neu expressions.Results Syn expression first increased and then decreased.Syn expression in the 6-12 h subgroup reached the peak.Compared to the control group,Syn expressions in the 0-2,2-6,and 6-12 h subgroups significantly increased(P<0.05).Compared to the 6-12 h subgroup,Syn expressions in the 12-24,24-48,and 48-72 h subgroups significantly decreased(P<0.05).Neu expression first decreased,then increased and decreased.Neu expression reached the trough in the 6-12 h subgroup and reached the peak in the 24-48 h subgroup.Compared to the control group and other subgroups,Neu expression significantly decreased(P<0.05)in the 6-12 h subgroup and significantly increased in the 24-48 h subgroup(P<0.05).Conclusion HI brain injury can affect the expressions of Syn and Neu.After injury,the brain nerves have strong synaptic and neuronal plasticity.
