摘要
目的探讨MYCT1通过PRSS21对喉癌细胞增殖和迁移的影响。方法应用实时PCR和Western blotting方法检测喉癌中PRSS21的表达水平及MYCT1对PRSS21表达水平的影响;应用CCK-8实验和克隆形成实验检测MYCT1通过PRSS21对喉癌细胞增殖和克隆形成能力的影响;应用划痕实验检测MYCT1通过PRSS21对喉癌细胞迁移能力的影响。结果PRSS21在喉癌中高表达(P<0.05);与对照组相比,MYCT1显著降低PRSS21在喉癌细胞中mRNA和蛋白质的表达水平(P<0.05);MYCT1通过PRSS21显著抑制喉癌细胞的增殖和克隆形成(P<0.05);MYCT1通过PRSS21显著抑制喉癌细胞的迁移能力(P<0.05)。结论MYCT1可通过抑制PRSS21的表达抑制喉癌细胞的增殖和迁移。
Objective To investigate the effects of MYCT1 on the PRSS21-mediated proliferation and migration of laryngeal carcinoma cells.Methods Real-time PCR and Western blotting were used to detect PRSS21 mRNA and protein expression,respectively in the context of MYCT1 silencing or overexpression in laryngeal carcinoma.The CCK-8 and clone formation assays were applied to monitor the effects of MYCT1 on the PRSS21-mediated proliferation and survival of laryngeal cancer cells.A scratch assay was further conducted to monitor the effect of MYCT1 on the migration(PRSS21-mediated)of laryngeal cancer cells.Results PRSS21 was highly expressed in laryngeal carcinoma(P<0.05).Compared to controls,MYCT1 significantly inhibited the expression of PRSS21 both at mRNA and protein levels in laryngeal cancer cells(P<0.05).Importantly MYCT1 significantly inhibited the proliferation,clone formation,and migration of laryngeal carcinoma cells,in a PRSS21 dependent fashion(P<0.05).Conclusion MYCT1 represses the proliferation and migration of laryngeal carcinoma cells via the inhibition of PRSS21 expression.
