基于Miseq测序分析海南阿宽蕉居群根际土壤细菌群落多样性

Analysis the Diversity of Bacterial Community from M. itinerans Population Rhizosphere Soil in Hainan Using Miseq Sequencing

摘要 阿宽蕉为热带雨林次生林中的先锋植物,由于克隆生长势强极易形成大面积的野生居群。为了解阿宽蕉根际土壤细菌多样性,本研究采用Illumina MiSeq高通量测序技术测定海南分布的8个野生阿宽蕉居群根际土壤细菌的16S r RNA基因的V3~V4变异区序列,分析不同居群土壤细菌群落结构。结果表明,阿宽蕉根际土壤细菌在97%的相似水平下共产生了14655个OTUs;产生OTU最多的样品是Xuxx21,为2324个;产生OTU最少的样品是Xuxx27,为1106个。分类学分析结果显示,8个样品共检测到25个菌门,96个菌纲,212个菌目,398个菌科,768个菌属,其中变形菌门和放线菌门为优势菌门。样品Xuxx25的Chao1指数为3036,香农指数为8.23,细菌菌群的丰度和多样性最高。样品Xuxx26和样品Xuxx27的细菌群落结构相似度最高。进一步分析表明,在8个样品中均发现纤维素降解复合菌系的关键功能属Clostridium,且在样品Xuxx24中的比例高达0.33%。本研究的结果揭示阿宽蕉根际土壤的细菌的群落多样性,为该生境的微生物资源开发提供依据。 Musa itinerans Cheesman is a pioneer in the secondary forest of tropical rainforest,which can easily form a large wild population due to its strong clonal growth.In order to understand the bacterial diversity of soil of M.itinerans,IIllumina Mi Seq high-throughput sequencing was used to determine the V3~V4 variation region of16 S r RNA gene in 8 populations of rhizosphere soil of M.itierans in Hainan,and the structure of bacterial communities in different populations was analyzed.The results showed that 8 rhizosphere soils in M.itinerans produced a total of 14655 OTUs at similar levels of 97%;the highest OTU yield was found from Xuxx21 at 2324;the lowest OTU yield was sample Xuxx27 at 1106.The results of the class analysis showed that 25 phyla,96 classes,212 orders,398 families and 768 genus were detected in 8 samples,of which Proteobacteria and Actinobacteria were the dominant floras.The sample Xuxx25 owed a Chao1 index of 3036 and a Shannon index of 8.23,with the highest abundance and diversity of bacterial flora.The sample Xuxx26 and sample Xuxx27 shared th e highest similarity of bacterial community structure.Further analysis showed that Clostridium,a key functional component of the cellulose-degrading bacterial genus,was found in all 8 samples,with a high proportion of 0.33%in the sample Xuxx24.This study revealed the diversity of bacteria in the rhizospheric soil of M.itinerans and provided basis for the exploitation of microbial resources from this habitat.
