摘要
为了考察抗阻运动对抑郁动物模型学习记忆能力的改善作用及机制。本研究建立了慢性温和不可预见性应激(CUMS)抑郁大鼠模型,通过对CUMS大鼠进行8周的抗阻运动,且每周2次向大鼠静脉注射0.1 mg/kg的ERK抑制剂PD98059,考察了大鼠的行为学、海马神经元凋亡及海马中Bcl-2、GAP-43、p-ERK和BDNF的表达。Open-field测试结果显示:抗阻运动提高了CUMS大鼠的水平运动次数和垂直运动次数(p<0.05);Morris水迷宫测试结果表明,抗阻运动减少了CUMS大鼠的逃避潜伏期,而增加了穿越平台次数(p<0.05);流式细胞术发现抗阻运动抑制了CUMS大鼠的海马神经元凋亡(p<0.05)。免疫组织化学染色显示,抗阻运动升高了海马区Bcl-2和GAP-43的表达(p<0.05)。蛋白质印迹结果显示,抗阻运动升高了海马区p-ERK和BDNF的蛋白表达(p<0.05)。而PD98059处理可逆转抗阻运动对CUMS大鼠的抗抑郁作用(p<0.05)。本研究结果表明,抗阻运动可提高CUMS大鼠的学习记忆能力并抑制海马区神经元凋亡,抗阻运动通过激活ERK信号通路来发挥抗抑郁作用。
In order to investigate the improvement effect and mechanism of resistance exercise on learning and memory ability of animal models of depression.In this study,a chronic unpredictable mild stress(CUMS)depression rat model was established.After modeling,CUMS rats underwent resistance exercise for 8 weeks and the rats were intravenously injected with 0.1 mg/kg ERK inhibitor PD98059 twice a week.Then the behavior of rats,hippocampal neuron apoptosis and the expression of Bcl-2,GAP-43,p-ERK and BDNF in hippocampus were investigated.Open-field tests showed that resistance exercise increased the number of horizontal and vertical exercises of CUMS rats(p<0.05).The Morris water maze test showed that resistance exercise reduced the escape latency of CUMS rats and increased the number of crossing platforms(p<0.05).Flow cytometry showed that resistance exercise inhibited hippocampal neuronal apoptosis in CUMS rats(p<0.05).Immunohistochemical staining showed that resistance exercise increased the expression of Bcl-2 and GAP-43 in the hippocampus(p<0.05).Western blotting results showed that resistance exercise increased the protein expression of p-ERK and BDNF in the hippocampus(p<0.05).The PD98059 treatment reversed the antidepressant effect of resistance exercise on CUMS rats(p<0.05).This study showed that resistance exercise can improve the learning and memory ability of CUMS rats and inhibit neuronal apoptosis in the hippocampus.Resistance exercise exerts antidepressant effects by activating ERK signaling pathway.
