艾拉莫德抑制细胞因子促进骨髓瘤RPMI-8226细胞的凋亡 被引量:2

Iguratimod Promotes Apoptosis of Human Multiple Myeloma RPMI-8226Cells by Inhibiting Cytokines

摘要 为了探讨艾拉莫德对人骨髓瘤细胞的增殖和凋亡的影响及其可能的作用机制。本研究通过CCK8和流式细胞术分别检测25μg/mL、50μg/mL和100μg/mL浓度艾拉莫德对人骨髓瘤RPMI-8226细胞增殖和凋亡的影响,ELISA法检测不同浓度的艾拉莫德对人骨髓瘤RPMI-8226细胞分泌细胞因子TGF-β和IL-6水平的影响,并通过Western blotting检测艾拉莫德对TGF-β、IL-6、Cleaved caspase 3和Bcl2蛋白表达的影响。艾拉莫德能够抑制人骨髓瘤细胞RPMI-8226细胞活力,呈剂量依赖性(p<0.05)。与对照组(0μg/mL)的细胞凋亡率(7.43±0.23)%比较,25μg/mL、50μg/mL、100μg/mL的艾拉莫德促进骨髓瘤RPMI-8226细胞凋亡率分别为(15.23±0.38)%、(31.70±0.31)%和(43.60±0.42)%,差异具有统计学意义(p<0.05)。ELISA检测结果显示,艾拉莫德能够显著抑制骨髓瘤RPMI-8226细胞因子TGF-β和IL-6分泌量(p<0.05)。Western blotting结果显示,艾拉莫德能够显著抑制骨髓瘤RPMI-8226细胞IL-6、TGF-β和Bcl2蛋白表达量显著升高,促进Cleaved caspase 3蛋白表达水平。艾拉莫德能够通过抑制细胞因子TGF-β和IL-6的表达水平,激活线粒体凋亡通路,促进骨髓瘤RPMI-8226细胞凋亡。 To investigate the effect of Iguratimod on proliferation and apoptosis of human multiple myeloma cells and its possible mechanism.The effects of iguratimod at the concentration of 25μg/mL,50μg/mL,100μg/mL on the viability and apoptosis of human myeloma RPMI-8226 cells were detected by CCK8 and flow cytometry.The effect of iguratimod on the secretion of cytokines TGF-βand IL-6 in human myeloma RPMI-8226 cells was detected by ELISA.The effect of iguratimod on the expression of TGF-β,IL-6,cleaved caspase 3 and Bcl2 in myeloma RPMI-8226 cells was detected by Western blotting.Iguratimod could inhibit the viability of human myeloma cell line RPMI-8226 in a dose-dependent manner(p<0.05).Compared with the apoptosis rate of the control group(7.43±0.23)%,the apoptotic rates of myeloma RPMI-8226 cells at the concentrations of 25μg/mL,50μg/mL,and 100μg/mL were(15.23±0.38)%,(31.70±0.31)%and(43.60±0.42)%,the difference was statistically significant(p<0.05).ELISA results showed that iguratimod could significantly inhibit the secretion of TGF-βand IL-6 in myeloma RPMI-8226(p<0.05).Western blotting results showed that irammod could significantly inhibit the expression of IL-6,TGF-βand Bcl2 in myeloma RPMI-8226 cells,and promote the expression of cleaved caspase 3 protein.Iguratimod could activate mitochondrial apoptosis pathway and promote myeloma RPMI-8226 cell apoptosis by inhibiting the levels of cytokines TGF-βand IL-6.
