摘要
为了揭示芎归汤结合跑台运动对大鼠冠心病治疗作用。将30只SD大鼠随机分为对照组、冠心病组和芎归汤+跑台运动组(n=10)。冠心病组和芎归汤+跑台运动组大鼠建立了冠心病模型。芎归汤+跑台运动组大鼠建模后进行芎归汤(1.5 g/kg)结合跑台运动(15 m/min,30 min/d)治疗,共治疗4周。考察各组大鼠的心功能和内皮功能指标。采用RT-PCR或Western blotting检测NLPR3、ET-1和v WF的表达。通过苏木素碱性品红苦味酸(HBFP)染色检测心肌微梗塞区域。使用TUNEL染色检测心肌细胞凋亡。研究显示,与冠心病组相比,芎归汤+跑台运动组大鼠的左室舒张末内径(LVEDD)和左室收缩末内径(LVESD)均降低,而左室射血分数(LVEF)升高(p<0.05)。与冠心病组相比,芎归汤+跑台运动组的心肌微梗塞区域和心肌细胞凋亡率均降低(p<0.05)。与冠心病组相比,芎归汤+跑台运动组大鼠的ET-1、v WF和NLRP3的m RNA和蛋白表达水平均降低(p<0.05)。芎归汤结合跑台运动通过抑制心肌细胞凋亡、ET-1、v WF、NLRP3炎性体通路显著提高了冠心病大鼠的心功能和内皮功能。
In order to reveal the therapeutic effect of Xionggui decoction combined with treadmill exercise on coronary heart disease in rats.Thirty SD rats were randomly divided into control group,coronary heart disease group and Xionggui decoction+treadmill exercise group(n=10).Coronary heart disease models were established in the coronary heart disease group and the Xionggui decoction+treadmill exercise group.The rats in the Xionggui decoction+treadmill exercise group were treated with Xionggui decoction(1.5 g/kg)combined with treadmill exercise(15 m/min,30 min/d)for 4 weeks.The cardiac and endothelial function indexes of rats in each group were investigated.RT-PCR or Western blot were used to detect the expression of NLPR3,ET-1 and v WF.The region of myocardial micro-infarction was detected by hematoxylin basic fuchsin picric acid(HBFP)staining.TUNEL staining was used to detect cardiomyocyte apoptosis.Studies showed that,compared with the coronary heart disease group,the LVEDD and LVESD of the Xionggui decoction+treadmill exercise group were reduced,while the LVEF was increased(p<0.05).Compared with the coronary heart disease group,the myocardial microinfarction area and the myocardial cell apoptosis rate in the Xionggui decoction+treadmill exercise group were both reduced(p<0.05).Compared with the coronary heart disease group,the m RNA and protein expressions of ET-1,v WF and NLRP3 in the Xionggui decoction+treadmill exercise group were reduced(p<0.05).Xionggui decoction combined with treadmill exercise significantly improved the cardiac and endothelial function of coronary heart disease rats by inhibiting cardiomyocyte apoptosis,ET-1,v WF and NLRP3 inflammatory body pathways.
