摘要
通过菠萝转录组测序分析,筛选到可能与开花和花序发育相关的基因AcSPL3(Ananas comosus squamosa promoter binding-like protein 3)。本研究克隆目的基因AcSPL3的cDNA序列,借助生物信息学和荧光定量技术对AcSPL3进行序列和表达特性分析并成功构建35S::AcSPL3过表达载体。研究结果表明AcSPL3含有SBP保守结构域和核定位信号,是植物特有转录因子SPL家族成员,AcSPL3在不同生长发育阶段的不同组织部位都表达,但表达量存在差异,并且从进化树中发现AcSPL3和OsSPL13在同一进化分支上。本研究为深入探究菠萝AcSPL3的表达模式及功能,研究AcSPL3在菠萝催花和花发育中的作用及菠萝开花机理提供参考依据。
Through analysis of pineapple transcriptome sequencing,genes AcSPL3(Ananas comosus squamosa promoter-binding-like protein 3)that may be related to flowering and flower development were screened.This study cloned the cDNA sequence of the target gene AcSPL3 and analyzed the sequence and expression characteristics of AcSPL3 with bioinformatics technology and quantitative real-time(RT-qPCR).In addition,we constructed 35 S::AcSPL3 overexpression vector successfully.The results showed that AcSPL3 contains SBP domain and nuclear localization signals and is a member of plant specific transcription factors SPLs.AcSPL3 was expressed in different tissues at different growth and development stages,but the expression levels were different.Furthermore,AcSPL3 and OsSPL13 are found on the same evolutionary branch from the evolutionary tree.This study provides a reference for further explore the expression pattern and function of pineapple AcSPL3,and studies on the role of AcSPL3 in flower development and flowering mechanism of pineapple.
