siRNA干扰CPEB4基因表达对胶质瘤侵袭与生长的影响 被引量:1

摘要 目的探讨胞质多聚腺苷酸化成分结合蛋白4(CPEB4)在胶质瘤发生发展过程中的作用。方法免疫组化、Western blot及Real Time PCR法检测胶质瘤组织和细胞中CPEB4的表达情况,Western blot及Real Time PCR法检测CPEB4 siRNA的干扰效率,裸鼠成瘤试验测定CPEB4基因表达下调后胶质瘤细胞在体内致瘤情况。结果免疫组化、Western blot和Real Time PCR结果显示脑胶质瘤组织CPEB4的蛋白和mRNA表达明显升高,随着WHO分级的增加,CPEB4的蛋白和mRNA水平也显著增加。Western blot和Real Time PCR结果显示U87细胞系CPEB4的蛋白和mRNA水平显著高于胶质瘤细胞系U251。在体干扰CPEB4表达能显著降低肿瘤的体积和质量。结论脑胶质瘤组织和细胞高表达CPEB4基因,在体干扰CPEB4表达能显著抑制脑胶质瘤细胞的生长,CPEB4可能成为治疗胶质瘤的重要靶点。 Objective To explore critical role of cytoplasmic polyadenylation element binding protein 4(CPEB4)in the development and progress of glioma.Methods Immunohistochemistry,Western blot and Real Time PCR were used to detect the expression of CPEB 4 in human glial tumor tissue and cells.The interference efficiency of CPEB4 siRNA was determined with Western blot and Real Time PCR.Tumor xenograft in nude mice was performed to investigate the effect of CPEB4 downregulation on the glioma growth in vivo.Results Immunohistochemistry,Western blot and Real Time PCR showed that the expression of CPEB4 protein and mRNA in brain glioma tissue was significantly increased.The protein and mRNA levels of CPEB4 were also significantly increased with the increase of WHO grade.The protein and mRNA levels of CPEB4 in U87 cell line were significantly higher than that of the glioma cell line U251.The volume and weight of tumors were significantly reduced by interference of CPEB4 in vivo.Conclusion The CPEB4 is highly expressed in glioma tissues and cells,and knockdown of CPEB4 inhibits the growth of glioma cells in vivo.CPEB4 may be an important target for the treatment of glioma.
