摘要
目的探讨PI3K/AKT/mTOR信号通路的单核苷酸多态性(SNPs)与胃癌发生、生存时间及病理特征的关联。方法采用PCR在PI3K、AKT、mTOR基因中鉴定PI3K rs7556371、rs6443624、rs7651265、AKT rs3803304、rs2498804、rs1130214、mTOR rs7211818和rs7212142多态性。应用非条件Logistic回归分析评估不同基因型与胃癌发病的风险、生存时间及病理特征的关系。结果与A等位基因相比,PI3K rs7651265 G等位基因与胃癌的发展显著相关(OR=4.913,95%CI 3.637-6.635,P<0.05)。不同年龄患者AKT rs1130214基因频率存在差异(P<0.05)。AKT rs3803304和rs1130214与胃癌的肿瘤浸润深度有关(P<0.05)。PI3K rs7651265与胃癌淋巴结转移有关(P<0.05)。AKT rs3803304与胃癌的分期有关(P<0.05)。结论研究结果表明PI3K rs7651265与胃癌风险显著相关,PI3K rs7651265 A等位基因与胃癌患者的生存时间有关,这些结果可能有助于胃癌的临床诊断和治疗。
Objective To investigate the association of single nucleotide polymorphisms(SNPs)in the PI3K/AKT/TOR signaling pathway with the occurrence,the survival time and the pathological features of gastric cancer.Methods Eight SNPs,PI3K rs7556371,rs6443624,rs7651265,AKT rs3803304,rs2498804,rs1130214,mTOR rs7211818 and rs7212142 in PI3K-AKT-mTOR gene,were genotyped by the polymerase chain reaction-ligase detection reaction method.Multivariate Logistic regression analysis was used to evaluate the relationships between different genotypes and the risk,the survival time and the pathological features of gastric cancer.Results The PI3K rs7651265 G was significantly associated with an increased gastric cancer risk compared with rs7651265 A(OR=4.913,95%CI:3.637-6.635,P<0.05).There was difference in the frequency of AKT rs1130214 among patients of different ages(P<0.05).The AKT rs3803304 and rs1130214 were associated with the tumor invasion depth in patients with gastric can-cer(P<0.05).PI3K rs7651265 was associated with lymph node metastasis(P<0.05),and AKT rs3803304 was related to the staging of gastric cancer(P<0.05).Conclusion The results suggest that PI3K rs7651265 polymorphism could affect the gastric cancer risk.The PI3K rs7651265 A allele can increase the survival rate of patients with gastric cancer.These results might contribute to the clinical diagnosis and treatment of gastric cancer.
