猪圆环病毒3型套式PCR检测方法的建立及应用

Establishment and Application of Nested PCR Method for Detecting Porcine Circovirus Type 3

为快速检测猪圆环病毒3型(PCV3),根据PCV3全基因组保守区域设计特异性引物,建立PCV3套式PCR检测方法,并对采自安徽省部分地区的528份猪临床样品进行检测。结果显示,建立的方法仅对PCV3检测阳性,最低检测限为1.74×10拷贝/μL,与商品化检测试剂盒的符合率高;临床样品中PCV3总阳性检出率为20.45%(108/528)。建立的PCV3套式PCR方法可用于PCV3的临床样品检测。

PCV3 infection exists in swine herds in most areas of Anhui.In order to rapidly detect porcine circovirus type 3(PCV3),specific primers were designed based on the conserved region of PCV3 genome,PCV3 nested PCR detection method was established,and 528 clinical samples collected from Anhui province were detected.The results showed that the detection method was only positive for PCV3,and the minimum detection limit was 1.74×10 copies/μL,which was consistent with the commercialization test kit;The total positive rate of PCV3 in clinical samples in Anhui was 20.45%(108/528).The established PCV3 nested PCR method is specific,sensitive,stable,economical and practical,and can be used for clinical detection of PCV3;PCV3 infection exists in pigs in most areas of Anhui.
