摘要
目的:建立不同产地首乌藤的指纹图谱,并测定首乌藤中没食子酸、反式二苯乙烯苷、大黄素的含量。方法:利用HPLC建立不同产地首乌藤的指纹图谱,并对12批首乌藤中没食子酸,反式二苯乙烯苷,大黄素进行定量分析。色谱条件为Extend-C18773450-902(4.6 mm×150 mm,5μm),以0.1%磷酸水(A相)-甲醇(B相)为流动相;梯度洗脱,流速:0.3 mL/min;检测波长:280 nm;柱温:30℃;进样量:5μL。结果:12批首乌藤相似度在0.915~0.994之间。没食子酸、反式二苯乙烯苷和大黄素的浓度分别在4~240μg/mL(R=0.999,8)、6.25~250μg/mL(R=0.999,9)、3~100μg/mL(R=0.999,8)内与峰面积呈良好的线性关系。结论:不同产地首乌藤中有效成分种类基本一致,但是含量差异较大。其中肝损成分二苯乙烯苷的含量远低于何首乌的含量,其临床用药的安全性好于何首乌。
Objective: To establish fingerprints of Shouwuteng(polygoni multiflori caulis) from different origins, and to determine the content of gallic acid, trans-stilbene glycoside and emodin in Shouwuteng(polygoni multiflori caulis). Methods: The fingerprints of Shouwuteng(polygoni multiflori caulis) from different origins were established by HPLC, and the gallic acid, trans-stilbene glycoside and emodin in 12 batches of Shouwuteng(polygoni multiflori caulis) were quantitatively analyzed. The chromatographic column was Extend-C18773450-902(4.6 mm×150 mm, 5 μm), with 0.1% phosphoric acid water(phase A)-methanol(phase B) as mobile phase;gradient elution, flow rate was 0.3 mL/min;detection wavelength was 280 nm;column temperature was 30 ℃;injection volume was 5 μL. Results: The similarity of 12 batches of Shouwuteng(polygoni multiflori caulis) ranged from 0.915 to 0.994. The concentration of gallic acid, trans-stilbene glucoside and emodin showed good linear relationship with peak area in the range of 4-240 μg/mL(R=0.999,8), 6.25-250 μg/mL(R=0.999,9), 3-100 μg/mL(R=0.999,8), respectively. Conclusion: The types of active ingredients in Shouwuteng(polygoni multiflori caulis) from different origins are similar, while the contents are quite different. Among them, the content of total stilbene glycosides in the liver damage component of Shouwuteng(polygoni multiflori caulis) is much lower than that of Heshouwu(Polygoni Multiflori Radix), and the safety of clinical drugs is better than that of Heshouwu(Polygoni Multiflori Radix).
