摘要
为研究RBP4基因单核苷酸多态性与贵州黑山羊产羔性状之间的关系,本研究采用混合DNA池测序和直接测序法筛选SNPs位点,进行生物信息学和群体遗传参数分析,并收集118头贵州黑山羊母羊1~4胎产羔数据,分析SNPs位点与产羔性状之间的关联性。结果显示:共检测到2个SNPs位点,为g.4527G>T和g.4744C>T。生物信息学分析发现g.4744C>T位点变异会使mRNA二级结构最小自由能增大,结构稳定性降低;群体遗传参数分析发现,g.4527G>T位点基因型频率差异较大,G为优势等位基因;g.4744C>T位点C为优势等位基因;χ2检验显示两位点均处于Hardy-Weinberg平衡。基因型与产羔数关联性分析发现,g.4527G>T位点在2、3、4胎次,野生型产羔数显著高于突变型;g.4744C>T位点在第1胎次,突变型母羊产羔数显著高于野生型。本研究揭示了RBP4基因对贵州黑山羊产羔数存在显著影响,g.4527G>T和g.4744C>T可能是影响贵州黑山羊产羔性状的重要SNPs位点。
In order to research the relationship between single nucleotide polymorphism of RBP4 gene and lambing traits of Guizhou black goat.In this study,SNPs were screened by mixed DNA pool sequencing and direct sequencing,collect the 1-4 litter data of 118 Guizhou black goat ewes,and analyze the correlation between SNPs sites and litter traits.Results two SNPs were detected,g.4527G>T and g.4744C>T.Bioinformatics analysis showed that g.4744C>T increased the minimum free energy of the secondary structure and decreased the structural stability.The analysis of population genetic parameters showed that the genotype frequency of g.4527G>T was quite different,G was the dominant allele.g.4744C>T locus C is the dominant allele.Theχ2 test showed that both locus were in Hardy-Weinberg equilibrium.The analysis of the relationship between genotype and lambing number showed that g.4527G>T locus,in the second,third and fourth parity,the lambing number of wild-type was significantly higher than that of mutant(P<0.05).For g.4744C>T locus,on the contrary,in the first parity,the lambing number of mutant ewe was significantly higher than that of wild type(P<0.05).This study revealed that RBP4 gene has a significant effect on the lambing number of Guizhou black goat.g.4527G>T and g.4744C>T may be the important SNPs loci that affect the lambing traits of Guizhou black goat.
