摘要
目的观察肿瘤坏死因子α诱导蛋白8样分子2(TIPE2)、叉头样转录因子3(Foxp3)和细胞毒性T淋巴细胞相关抗原4(CTLA-4)mRNA在原发性肝癌患者外周血单个核细胞中的表达及意义。方法前瞻性选择2019年1月至2019年12月至黄河三门峡医院进行原发性肝癌治疗的患者60例作为观察组。随机选择同一时间段至黄河三门峡医院进行体检的健康人员60人作为对照组。采用实时荧光定量(PCR)反应检测两组对象的TIPE2、Foxp3和CTLA-4mRNA的表达情况;比较观察组不同病理分期、有无转移灶TIPE2、Foxp3和CTLA-4mRNA表达情况差异;使用Pearson相关性分析,观察两组RIPE2、Foxp3和CTLA-4mRNA水平与AFP、γ-GT、GP-73水平的相关性。结果①观察组TIPE2mRNA水平低于对照组(0.57±0.09vs.0.71±0.12),Foxp3、CTLA-4mRNA水平高于对照组(0.70±0.23 vs.0.56±0.13、0.62±0.20vs.0.44±0.13),差异具有统计学意义(P<0.05)。②观察组不同肝癌分期患者TIPE2、Foxp3、CTLA-4mRNA水平差异具有统计学意义(P<0.05),Ⅱ、Ⅲ期患者TIPE2mRNA低于Ⅰ期(0.42±0.10vs.0.34±0.06vs.0.69±0.11),Foxp3、CTLA-4mRNA水平高于I期(0.72±0.28vs.1.23±0.37vs.0.59±0.21、0.59±0.13vs.0.98±0.25vs.0.37±0.06),Ⅲ期患者TIPE2mRNA低于Ⅱ期,Foxp3、CTLA-4mRNA水平高于II期,差异具有统计学意义(P<0.05)。③观察组中有转移灶患者TIPE2mRNA水平低于无转移灶患者(0.41±0.10vs.0.62±0.21),Foxp3、CTLA-4mRNA水平高于无转移灶患者(0.94±0.33vs.0.61±0.24、0.90±0.28vs.0.56±0.17),差异具有统计学意义(P<0.05)。④使用Pearson相关性分析,结果发现TIPE2与AFP呈负相关(r=-0.727,P<0.05),其余无相关性(P>0.05)。结论原发性肝癌患者的TIPE2、Foxp3和CTLA-4mRNA表达出现了异常,且在肿瘤发展的不同阶段均出现不同表达,因此考虑TIPE2、Foxp3和CTLA-4mRNA表达在肝癌的发生发展中起重要作用。
Objective To observe the expression and significance of tumor necrosis factor alpha-induced protein 8-like molecule 2 and forkhead-like transcription factor 3 mRNA in peripheral blood mononuclear cells(PBMC)of patients with primary liver cancer.Methods Randomly select 60 patients from January 2019 to December 2019 to Yellow River Sanmenxia Hospital for primary liver cancer treatment as the observation group.Randomly select 60 healthy people who will go to Yellow River Sanmenxia Hospital for medical examination at the same time period as a control group.The expressions of TIPE2,Foxp3 and CTLA-4 mRNA in the two groups were detected using real-time fluorescence quantitative(PCR)reaction method.The differences in the expression of TIPE2,Foxp3 and CTLA-4 mRNA in the observation group with different pathological stages,with or without metastasis were compared.The correlation between the two groups of RIPE2,Foxp3 and CTLA-4 mRNA levels and AFP,γ-GT,GP-73 levels were compared using Pearson correlation analysis.Results①The levels of TIPE2 mRNA in the observation group were lower than those in the control group(0.57±0.09 vs.0.71±0.12),and the levels of Foxp3 and CTLA-4 mRNA were higher than those in the control group(0.70±0.23 vs.0.56±0.13,0.62±0.20 vs.0.44±0.13),and there were statistical difference(P<0.05).②The levels of TIPE2,Foxp3 and CTLA-4 mRNA in the observation group were significantly different in patients with different liver cancer stages(P<0.05).TIPE2 mRNA in stageⅡandⅢpatients was lower than that in stageⅠ(0.42±0.10 vs.0.34±0.06 vs.0.69±0.11),Foxp3 and CTLA-4 mRNA levels were higher than stageⅠ(0.72±0.28 vs.1.23±0.37 vs.0.59±0.21,0.59±0.13 vs.0.98±0.25 vs.0.37±0.06),and TIPE2 mRNA in stageⅢpatients was lower than stageⅡ,Foxp3 and CTLA-4 mRNA levels were higher than stageⅡ,and the difference was statistically significant(P<0.05).③The patients with metastases in the observation group had lower TIPE2 mRNA levels than those without metastases(0.41±0.10 vs.0.62±0.21),Foxp3 and CTLA-4 mRNA levels were higher than those without metastases(0.94±0.33 vs.0.61±0.24,0.90±0.28 vs.0.56±0.17),and there were statistical differences(P<0.05).④Using Pearson correlation analysis,the results showed that TIPE2 and AFP were negatively correlated(P<0.05),and the rest were not correlated(P>0.05).Conclusion The expression of TIPE2,Foxp3 and CTLA-4 mRNA is abnormal in patients with primary liver cancer,and different expressions occur at different stages of tumor development.Therefore,TIPE2,Foxp3 and CTLA-4 mRNA expression are considered to play a role in the occurrence and development of liver cancer.To an important role.
