人参总皂苷对多囊卵巢综合征模型大鼠激素水平及NGF表达的影响 被引量:5

Effects of ginseng total saponins on hormone levels and NGF expression in rats with polycystic ovary syndrome

摘要 目的研究人参总皂苷(GTS)对多囊卵巢综合征(PCOS)模型大鼠的治疗作用。方法选取SPF级雌性SD大鼠,通过肌内注射4 mg脱氢表雄酮(DHEA)诱导PCOS模型,然后随机分为空白组(正常SD大鼠+腹腔注射等量等时长0.9%氯化钠溶液)、模型模型对照组(PCOS模型大鼠+腹腔注射等量等时长0.9%氯化钠溶液)和GTS干预组(PCOS模型大鼠+建模次日每隔1天腹腔注射GTS 50 mg/kg,连续28 d)。实验结束观察大鼠卵巢形态,并测定其血清中激素水平及卵巢、垂体和海马体中神经生长因子(NGF)水平。结果与空白组比较,模型对照组大鼠的卵巢呈多囊样改变,体重和卵巢质量均显著增大,游离睾酮(FT)、黄体生成素(LH)/促卵泡激素(FSH)水平及卵巢、海马体和垂体中NGF的表达均显著升高,差异有统计学意义(P<0.05);而与模型对照组相比,GTS组体重和卵巢质量均显著降低,FT、LH/FSH水平及卵巢中NGF中的表达均显著降低,差异有统计学意义(P<0.05)。结论GTS对PCOS模型大鼠有潜在的治疗作用。 Objective To investigate the effects of ginseng total saponins(GTS)on rat model of polycystic ovary syndrome(PCOS).Methods SPF female SD rats were selected to established,the PCOS model which was induced by intramuscular injection of 4 mg dehydroepiandrosterone(DHEA).Then they were randomly divided into a blank group(normal SD rats+intraperitoneal injection of the same amount of normal saline),model model control group(PCOS model rats+intraperitoneal injection of the same amount of normal saline)and GTS intervention group(PCOS model rats)+GTS 50 mg/kg was injected intraperitoneally every other day on the next day of modeling for 28 consecutive days).At the end of the experiment,the morphology of the rat's ovary was observed,and the hormone levels in the serum and the levels of nerve growth factor(NGF)in the ovary,pituitary and hippocampus were determined.Results As compared with the blank group,cyst-like follicles in ovary tissues of control group were observed,the weights of bodies and ovaries,the levels of FT,LH,FSH and expressions of NGF in ovaries,pituitary and hippocampus were all increased,the differences were statistically significant(P<0.05);compared with the model control group,the body mass and ovarian mass of the GTS group were significantly reduced,and the levels of FT,LH/FSH and the expression of NGF in ovary were significantly reduced,the differences were statistically significant(P<0.05).Conclusion GTS has a potential therapeutic effect on rats of PCOS.
